The Verbena plant Callicarpa nudiflora Hook.et Arn is a species belonging to the genus Callicarpa,and its medicinal part is the dry part of the ground.It is a kind of authentic medicinal material in Hainan,and it is also one of the commonly used herbs of Hainan Li nationality doctors.With hemostasis,anti-thrombosis,anti-inflammatory,antibacterial,cytotoxic activity,enhance immunity and other effects,the clinical is mainly used to anti-inflammatory hemostasis treatment.The dried leaves of the medicinal materials were crushed and extracted by refluxing with 80% ethanol,followed by extraction with solvents dichlorometh ane,ethyl acetate,and water-saturated n-butanol.Using silica gel,gel,C18 and other column chromatography and reversed phase medium pressure liquid chro matography,high performance liquid chromatography and other methods for se paration,the final extraction of 10 compounds from n-butanol.Using chemical spectroscopy,chemical methods and comparative literature,the chemical struct ures of 10 compounds were determined,including three kinds of iridoids:callicarpa nudiflora A1(1),callicarpa nudiflora A2(2),callicarpa nudiflora A3(3);four kinds of phenylpropanoids: verbascoside(4),islatrericin(5),6-O-caffeoy l-β-D-glucose(6),6-O-caffeoyl-α-D-Glucose(7);two kinds of flavonoids: luteolin(8),luteolin-4’-O-(6’’-E-caffeoyl)-β-D-glucopyranoside(9)and one kind of Triter penoid: 2α,3α,19α,23-tetrahydroxy-12-ene-28-usutic acid(10).compounds 1,2 and 3 are novel compounds.HPLC method was applied in this experiment,HPLC fingerprinting was used to establish the fingerprint of Callicarpa nudiflora Granules.Thirteen com mon peaks were established in the experiment and 7 components were identif ied using the reference method.In addition,the similarity analysis was perfor med using the chromatographic fingerprint similarity evaluation system of trad itional Chinese medicine(2012).The similarity was above 0.9 and the similar ity was high.In addition,by comparing the HPLC chromatogram of Callicarpa nudiflora Granules with that of 10 batches of Callicarpa nudiflora leaves and 10 batches of Callicarpa nudiflora branch HPLC chromatogram peak area and number of peaks,it is presumed that the raw material used in the preparation of Luo Hua Zi Zhu Keli was added to non-medicinal parts of the branch or during the production process,has an effect on the components of verbascoside and ismailaside,resulting in a difference between the granular preparation and the Callicarpa nudiflora leaves.The study of fingerprints has improved the quality control of Callicarpa nudiflora Granules preparations.The established method has strong specificity and high accuracy.The identification of Callicarpa nudiflora Granules can be used as a reference.On the other hand,for the study of callicarpanudiflorae components,mainly used for better development and utilization of callicarpanudiflorae,improve its medicinal value,and further analysis of its medicinal material base.The research on fingerprints will further improve the quality control,ensure the uniform and stable qualified medicines for the production quality of the pharmaceutical enterprises,and ensure people to have a safe level of medication.
