Callicarpa nudiflora Hook.et Arn.,a plant belonging to the family of Verbenaceae,is distributed in southern China,India and Malaya and is one of popular Li medicine in Hainan Province.The plants from the Callicarpa genus have been reported to exhibit a variety of biological effects,such as anti-inflammatory,haemostatic,antibacterial,cytotoxic and immuno-enhancing activities.Prophase chemical studies on this species have led to the isolation of diterpenoids,triterpenoids,flavonoids,phenolic acids,volatile oils and so on.By means of various chromatographic methods such as silica gel,opening ODS column chromatography,semi-preparative HPLC,26 compounds were isolated from water extract of the leaves of Callicarpa nudiflora Hook.et Arn.,and the structures of 21 compounds were elucidated on the basis of spectroscopic data and physico-chemical methods.We proposed the biosynthesis pathway of the diterpenoids compounds.The compounds comprised 13 new compounds,named Callicarpic acid N1(1),Callicarpic acid N2(2),Callicarpate N1(3),Callicarpic acid N3(4),Callicarpic acid N4(5),Callicarpic acid N5(6),Callicarpate N2(7),Callicarpic acid N6(8),Callicarpic acid N7(9),Callicarpic acid N8(10),Callicarpate N3(11),Callicarpate N4(12),Callicarpic acid N9(13).The other 8 known compounds were identified as 7α-hydroxy sandaracopimaric acid(14),(-)-syringaresinol(15),medioresinol(16),(7S,8R,7,S,8’R)-7-(3,4,5-trimethoxyphenyl)-6-(4’hydroxy-3’-methoxyphenyl)-10,10’-dioxabicyc-lo[3.3.0]octane(17),(+)-syringaresinol(18),5-hydroxy-3,7,3’,4’-tetramethoxyflavone(19),5,4’-dihydroxy-3,7,3’-trimethoxyflavone(20),ursolic acid(21).The anti-inflammatory activities of compounds 1-21 were tested.Compounds 1-18 exerted inhibitory activity against the production of nitric oxide(NO)in RAW264.7 stimulated by lipopolysaccharide(LPS).However,compounds 20 and 21 showed cytotoxic activity.
