Objective To investigate the relationship between cognitive dysfunction and inflammatory response in hippocampus of middle aged mice after operation and the pretreatment effect of electro-acupuncture on postoperative cognitive dysfunction(POCD)by modulating the inflammatory response.Methods 216 male healthy clean level aged mice(18 to 20 months),in accordance with the principle of completely randomized,were divided into older control group(group OC),older model group(group OM),older drug group(group OD),and the electroacupuncture group(group EA),(n=54);54 healthy clean level adult male mice(2 months)were divided into control group(group YC)(n=54).Each group can be divided into three subgroups upon their execution time:1 day(1D),3 days(3D),7 days(7D)(n=18).All rats tested Morris water maze(MWM)navigation experiment in preoperative five consecutive mornings,and accepted their corresponding intervention and balance treatments in the afternoon that EA group received EA intervention with the points of "baihui","dazhui",D group were treated with minocycline(40 mg/kg)by intraperitoneal injection.The rest were fixed and intraperitoneal injection of saline solution to balance the processing factors.Control group were used for blank comparison.On the 6th day morning,mice were taken a hepatectomy to establish POCD model under isoflurane inhalation anesthesia.Each group taken MWM space exploration experiment and reversal test,O maze experiment,open filed test after 1d,3d,7d after operation respectively,then executed to death.Observation indications:(1)Detectd spatial learning ability(swimming distance)and spatial memory function(the original crossing number)by Morris water maze;(2)using O maze to test anxiety of mice(residence time and distance in open arms)(3)using enzyme-linked Immunosorbent(ELISA)to measure IL-1β,IL-6,TNF-α,HMGB1,VCAM-1,ICAM-1 in serum;(4)using ELISA to detect IL-6,IL-1β,TNF-a,HMGB1,Ach,VCAM-1,ICAM-1 in hippocampus..Results The first part 1.Morris water maze navigation laboratory test results:YC group compared with the same time,the OC group swimming distance increased significantly(P<0.01).Morris water maze space exploration experiment results:compared with YC group,OC group in the platform quadrant residence time shorten(P<0.01).O maze test results:compared with YC group,OC group stay open arm cut short time percentage(P<0.01).Serum IL-1β,IL-6,TNF-α,HMGB1 expression level,ICAM-1,VCAM-1 test results:compared with YC group,serum IL-1βOC group beta,IL-6,TNF-α,HMGB1,ICAM-1,VCAM-1 expression increased(P<0.01).The second part 2.Morris water maze navigation experiment results:all on the first day in aging mice swimming distance difference has no statistical significance(P>0.05).OD,EA group from day 3 to 4 days compared with OC,OM group swimming distance at the same time,the difference was statistically significant(P<0.01,P<0.05).Morris water maze space exploration experiment results:compared with the OC group at the same time,OM,OD1d,EA1d,cut short staying time in the target quadrant EA3d group,the difference was statistically significant(P<0.01,P<0.05);OM group compared with the same time,OD group and EA group target quadrant residence time growth,the difference was statistically significant(P<0.01);OD group compared with the same time,residence time in the target quadrant shorten EAld group,the difference was statistically significant(P<0.05).O maze test results:compared with the OC group at the same time,OM,OD1d,EAld,EA3d group cut short residence time in the open arms percentage,the difference was statistically significant(P<0.01,P<0.05);OM group compared with the same time,residence time percentage OD group and EA group open arms shorter growth,the difference was statistically significant(P<0.01);Compared with same time OD group,EA1d group cut short residence time in the open arms percentage,the difference was statistically significant(P<0.05).Elderly group mice serum and hippocampus in beta,IL-6,IL-1 TNF-α,HMGB1 expression level test results:compared with the OC group at the same time,OM,OD1d,EAld,EA3d expression was significantly increased,the difference was statistically significant(P<0.01,P<0.05);OM group compared with the same time,OD group and EA group decreased,the difference was statistically significant(P<0.01);Compared with group at the same time OD,EA1d group expression increased,the difference was statistically significant(P<0.05).Elderly group mice hippocampus Ach expression level test results:compared with the OC group at the same time,OM,OD1d,EAld,expression significantly reduce EA3d group,the difference was statistically significant(P<0.01,P<0.05);OM group compared with the same time,OD group and EA group,the difference was statistically significant(P<0.01);OD group compared with the same time,express EA1d group reduced,the difference was statistically significant(P<0.05).In the elderly group mice serum and hippocampus ICAM-1,VCAM-1:expression level test result and OC group comparison at the same time,OM,OD1d,EA1d,EA3d expression was significantly increased,the difference was statistically significant(P<0.01,P<0.05);OM group compared with the same time,OD group and EA group decreased,the difference was statistically significant(P<0.01);Compared with group at the same time OD,EA1d group expression increased,the difference was statistically significant(P<0.05).Conclusion 1.With aging,the ability of learning and memory in mice and inflamematory factor expression,is likely to be older POCD mice susceptible to base;2.Lobe resection has led to an increased pro inflammatory cytokines,resulting in hippocampal inflamemation,may be one of the mechanism of senile mice POCD;3.Postoperative peripheral electric acupuncture preconditioning inhibits senile mice and the expression of inflammatory cytokines in the hippocampus,probably is one of the older mice POCD intervention methods.
