| Backgroud and ObjectivePostoperative cognitive dysfunction((Postoperative cognitive dysfunction,POCD)is a common central nervous system complication in elderly patients after anesthesia and surgery,which is mainly characterized by confusion in consciousness,cognition,orientation,thinking and memory.At present,the main mechanism,effective treatment and measures are not clear.Studies have confirmed that synaptic proteins are closely related to learning and memory.Guhan YangshengJing is a key protected variety of proprietary Chinese medicine in China,which has the effects of preserving health and strengthening the brain,and its main components,such as ginseng and epimedium,have a protective effect on the nervous system.Therefore,after the aged rats were pretreated with Guhan YangshengJing gastric perfusion for 30 days,we performed laparotomy under isoflurane inhalation anesthesia to establish the animal model of postoperative cognitive impairment.Toexplore whether Guhan YangshengJing can improve postoperative cognitive dysfunction and whether it is related to synaptic proteins.MethodsThe 48 aged sprague-dawley(SD)rats(20-22 months old)were randomly divided into 4 groups: the control group,Guhan Yangshengjing1.6g/kg,the operation group,the operation group + Guhan Yangshengjing1.6g/kg.The control group and operation group were given the same amount of normal distilled water for 30 days.POCD model was established by exploratory laparotomy under isoflurane inhalation anesthesia.The Morris water maze test was performed 3 days after operation to detect the swimming route,latency,times of crossing the original platform and target quadrant time ratio of the aged rats after operation.The levels of Syn-1 and PSD-95 in hippocampus were detected by Western Blot.Results1.The water maze test showed that the swimming route of the aged rats in the operation group was complicated,the latency of searching for the platform was prolonged 3 days after operation,and the times of crossing the platform and the time ratio of target quadrant were significantly reduced.After 30 days of pretreatment by Guhan Yangshengjing(1.6g/kg/d,i.g.),the swimming route of the mice was simple,the latency of the platform was shortened,and the times of crossing the platform and the time of target quadrant were increased.2.The WB test results showed that the expression of SYN-1 and PSD-95 in hippocampus of aged rats in the surgery group decreased on the 7th day after surgery,and the.Guhan Yangshengjing(1.6g/kg/d ig)was given continuous gastric pretreatment for 30 days.The group can significantly increase the expression levels of synaptophysin SYN-1 and PSD-95 in hippocampus of elderly rats after surgery,and the difference is statistically significant.The expression levels of synaptic proteins SYN-1and PSD-95 in the control group,the control group + Guhan Yangshengjing(1.6g/kg/d ig)group and the surgery + Guhan Yangshengjing(1.6g/kg/d ig)group have no obvious difference.ConclusionThe improvement of cognitive impairment in elderly rats by Guhan Yangshengjing may be related to the up-regulation of synaptic proteins SYN-1 and PSD-95 in the hippocampus. |
