[Backgroud and Objective]Cognitive dysfunction often occurs in elderly patients with different degrees of cognitive dysfunction after anesthetic surgery,but the specific pathogenesis and effective methods of prevention and treatment are still not clear.The inflammatory response of hippocampus in the brain participates in the formation of learning and memory dysfunction.Guhan Yangshengjing has a protective effect on brain tissue.Its main component,ginseng,has a certain regulatory effect on the central nervous system,which can enhance learning and memory and improve cognitive function.Therefore,we used 20-22 months old rats to pre-treat rats with Guhan Yangshengjing oral liquid for 30 days.Under isoflurane anesthesia,laparotomy was performed to establish an animal model of postoperative cognitive dysfunction.To explore whether Guhan Yangshengjing can improve postoperative cognitive dysfunction in aged rats,and whether its mechanism is related to improving the inflammatory response in the hippocampus of rats.[Methods]105 Sprague-Dawley(SD)male rats aged 20-22 months were randomly divided into 5 groups:Control、Surgery、Guhan Yangshengjing 0.8g/kg+ Surgery、Guhan Yangshengjing 1.6g/kg+ Surgery、Guhan Yangshengjing 3.2g/kg+ Surgery,21 per group.Continuous intragastric pretreatment of Guhan Yangshengjing for 30 days,the control group and the surgery group were treated with distilled water of equal volume for 30 days.Then the POCD rat model was established by laparotomy with isoflurane anesthesia.The Y-maze and novel object recognition experiments were performed 3 days after surgery to detect the cognitive function of the rats.Detection of IL-6 and TNF-alpha in hippocampus of rats by Western Blot.[Results]1.Exploratory laparotomy with isoflurane anesthesia in 20-22 months old rats,Behavioral Y-maze and novel object recognition experiments were performed 3 days after operation.The results showed that: In the Y-maze experiment,the rate of alternation of the aged rats was significantly reduced,but Guhan Yangshengjing(1.6 and 3.2g/kg/d,i.g.)pretreatment for 30 days can significantly increase the alternation accuracy of POCD rats;In the novel object recognition experiment,the novel object recognition index of postoperative elderly rats was significantly reduced,however,the Guhan Yangshengjing(1.6 and 3.2g/kg/d,i.g.)pretreatment for 30 days can significantly increase the novel object recognition index of POCD rats.The above results indicate that cognitive dysfunction occurs in postoperative aged rats,and Guhan Yangshengjing can improve the postoperative cognitive dysfunction of elderly rats.2.WB was used to detect the changes in the expression levels of proinflammatory cytokines in the hippocampus of aged rats at different time points after surgery,and the results showed that on the 1st and 3rd postoperative day,the expression of proinflammatory cytokines TNF-a and IL-6 in the hippocampus of aged rats increased significantly,while Guhan Yangshengjing(1.6g/kg/d,ig)pretreatment 30 days can significantly reduce the levels of pro-inflammatory cytokines TNF-a and IL-6 in hippocampus of postoperative aged rats,indicating that the inflammatory response of hippocampus in aged rats is increased.Guhan Yangshengjing can improve inflammation reaction in hippocampus of postoperative aged rats.[Conclusion]Guhan Yangshengjing can improve cognitive dysfunction in postoperative aged rats,and its mechanism is closely related to improving inflammatory reaction in hippocampus of postoperative elderly rats.
