Objective: To understand the expression of Toll-like receptor 2(TLR2)and Toll-like receptor 4(TLR4)in myeloid dendritic cells(mDC)and plasmacytoid dendritic cells(pDC)in the peripheral blood of patients with systemic lupus erythematosus(SLE),and to explore the role of both in the pathogenesis of SLE and their clinical significance.Methods: 54 SLE patients who met the enrollment criteria were selected as the study subjects,and the SLE patients were divided into stable group(≤4 points)and active group(≥5 points)according to the SLEDAI score,while 30 healthy volunteers from our hospital were selected as the control group,and flow cytometry(FCM)was applied to detect the expression of TLR2 and TLR4 on the surface of mDC and pDC in the peripheral blood of SLE patients and healthy volunteers.The expression of TLR2 and TLR4 on the surface of mDC and pDC in peripheral blood of SLE patients and healthy volunteers was measured by flow cytometry(FCM).The expression levels of TLR2 and TLR4 on the surface of mDC and pDC in the peripheral blood of SLE patients and healthy controls,and the relationship between them and the relevant clinical indexes were analyzed with the help of statistical software.Results:(1)the expression rate of mDC-TLR2 was significantly higher in the SLE active group than in the stable group(P<0.01)and the healthy control group(P<0.01);the expression rates of pDC-TLR2,mDC-TLR4 and pDC-TLR4 were significantly higher in the SLE active group than in the healthy control group(P<0.05);(2)the expression rate of mDC-TLR2 was negatively correlated with serum Ig G level and24 h urine protein quantification(P<0.05);the expression rates of mDC-TLR4 and pDC-TLR4 were both positively correlated with platelet count(P<0.05);the expression rate of pDC-TLR4 was significantly negatively correlated with 25-OH-VD(P<0.05).Conclusions:(1)The expression rates of TLR2 and TLR4 on the surface of mDC and pDC were significantly increased in SLE patients,suggesting that TLR2 and TLR4 may play an important role in the pathogenesis of SLE;(2)the expression rate of mDC-TLR2 correlated with serum Ig G level and 24 h urine protein quantification and other common clinical indicators;(3)Serum 25-OH-VD deficiency may have an effect on the expression of TLR4 on the surface of pDC.
