| Objective:Research the expression of Toll-like receptor9 (TLR9) in PBMCs and the expression of interferon-alpha (IFN-α) and interleukin 10(IL-10) in serum with SLE patients; Analysis the correlation of TLR9,IFN-αand IL-10 with SLEDAI;Analysis the correlation between IFN-αand IL-10. Explore their roles in the pathogenesis of SLE and provide new ideas for the clinical treatment.Methods:To segregate PBMCs from 60 cases of SLE patients and 40 healthy controls.Detect the expression of TLR9 mRNA in PBMCs by real-time quantitative polymerase chain reaction(Real-time Q-PCR) with SLE patients and healthy controls; Detect the expression of IFN-αand IL-10 by enzyme-linked immunosorbent assay (ELISA) with SLE patients and healthy controls; Analysis the correlation of TLR9,IFN-αand IL-10 with SLEDAI by Pearson or Spearman. Analysis the correlation between IFN-αand IL-10 by Person or Spearman;Results:1. Real-time Q-PCR results showed that SLE group and control group were express Toll-like receptor9 (TLR9); the expression of TLR9 mRNA in activity group significantly higher than inactivity group and healthy controls, the difference was statistically significant(P<0.01); but their have no statistical significance.between inactivity group and healthy controls (P>0.05).2. ELISA test results showed that the expression of IFN-αand IL-10 in activity group was significantly higher than inactivity group, the difference was statistically significant(P<0.01)); And the two groups were significantly higher than the healthy group, the difference was statistically significant(P<0.01). 3.Correlation analysis showed that:the levels of TLR9,IFN-αand IL-10 were positively correlated with SLEDAI(P<0.01); the levels between IFN-αand IL-10 were also positively correlated(P<0.01)).Conclusion:1. The expression of TLR9 mRNA in activity group significantly higher than inactivity group and healthy controls, the difference was statistically significant; but their have no statistical significance.between inactivity group and healthy controls.It indicate that TLR9 play a a certain significance role.in the pathogenesis of SLE.2. The expression of IFN-αand IL-10 in SLE group was significantly higher than healthy controls, the difference was statistically significant; It indicate that Tr function with SLE patients of IFN-αand IL-10 secretion increased.3. The levels of TLR9,IFN-αand IL-10 with SLEDAI were positively correlated;so we can detect the SLE activites by TLR9,IFN-αand IL-10.4. The expression between IFN-αand IL-10 were positively correlated., it suggested the interaction between cytokines. |
