The coldness and drought are the important factors limiting grape production,especially in my country’s northeast,North China and Northwest China.In recent years,Ningxia in my country has developed into my country’s main brewing grape feeding areas,how to choose cold grapes,reduce the cost of cold-resistant and cold,and use exposure to plant or slightly buried cold and cold,is Ningxia grape required main issues facing other similar production areas in my country.This study was based on the five strains of Chinese Wild Vitis piasezkii Maxim and Vitis riparis Michx,Beibinghong,Cabernet Sauvignon,Zuoyouhong,Beichun,red globe.The relative conductivity、POD、SOD、CAT、malondialdehyde and proline content were determination.through membership function,and identified the cold resistance of 11 grape varieties or strains.The main results are as follows:1.Using the cold-resistant drought-resistant correlation index to identify the cold resistance of grapes: Get the trend of the physiological changes of grape branches under cold stress: With the intensification of cold stress,the relative conductivity of the branches has gradually risen,and the better cold resistance,The slower rise,the lower the electrical conductivity;the anti-oxidase activity of the branches will presen a tendency to increase after raising the temperature,and the better the cold resistance,the lower the temperature of the oxidase active peak,and the larger the peak value;The propylenedehyde content continues to rise as the temperature is reduced,the lower the propylenedehyde content,the better the cold resistance;the proline content increases with cold stress,and the trend is preceded and the value is large.The better the coldness.2,According to the affiliate function,the coldness of the grape plant strains is evaluated: the 11 material branches of the test materials are evaluated by cold-resistant identification,Beichun,Beibinghong,and Zuoyouhong belong to high cold-resistant varieties.Originally,the river bank grapes in the United States belong to high cold-resistant varieties,and European grape varieties ’Cabernet Sauvignon’ belongs to the low cold-resistant variety,and the red globe belongs to susceptible resistance varieties.The Guyuan-1 is a high cold-resistant strain;Gansu-91 belongs to the medium cold-resistant strain,and the Baishui-40 belongs to the low cold-resistant strain.Huaxian-2 and the Liuba-9 belong to susceptible resistance strain.3.This study was determined by real-time fluorescent PCR to determine the river bank grape,Beibinghong,Guyuan-1,and Cabernet Sauvignon ’s refrigerated gene Vv CBF1 in different low temperatures.As the temperature decreases,the gene expression of the Beibinghong varieties is the highest,followed by the river bank grape,Guyuan-1,Cabernet Sauvignon.The expression of Cabernet Sauvignon Vv CBF1 showed a trend of first decreasing,then increasing and then decreasing with the decrease of temperature,there was a significant increase in controls at other temperatures;the Guyuan-1 expression increased at-8°C and-16 °C,no Significant differences,expression of expression at-20 °C and 24 °C also have no significant difference;the expression of the Beibinghong Vv CBF1 is compared to the control,and there is no significant difference in the control,and there is no significant difference in other temperatures,and the overall trend is presented first decreases.The rear rising trend is similar to the antioxidant,proline trend;the expression of river bank Vv CBF1 gene is more stable,and there is no significant difference in expression of 4 °C and-16 °C,and there is also no significant difference in other temperatures.The high and low sorting of the four materials Vv CBF1 gene expression is consistent with comprehensive evaluation.Therefore,the expression of Vv CBF1 genes can be used as auxiliary proof of the cold physiological indicators.4.In addition,in the control of the water supply 0d,measurement of POD,SOD,CAT activity and the change of proline content,obtained in the water 9d,4 grape material blade antioxidant activity and proline content have a continuous rise.According to the membership function,four physiological indicators are comprehensively drought resistant,and beta is a high drought-resistant variety,but the Guyuan-1,Beibinghong,Beichun are medium drought-resistant varieties or strains.In summary,based on the test material coldness identification,the identification and analysis of cold resistance and drought resistance,the expression analysis of the cold transcription factor gene VVCBF1 is obtained,and the study has been obtained from Chinese Wild Vitis piasezkii Maxim.Guyuan-1 belongs to high cold-resistance,medium-resistant strain,can serve as a cold-resistant breeding application,Gansu-91 belongs to cold-resistance strain,Baishui-40 belongs to the low cold-resistance strain,Huaxian-2 and Liuba-9 belong to the susceptible resistance strain;Beibinghong and Zuoyouhong varieties can be cultivated in Ningxia in the periodic cold year,as a anti-cold protective cultivation to reduce the cold loss.
