| Objective: In order to provide scientific basis for the clinical treatment of Qixian Tongluo Formula,the effect and mechanism of Qixian Tongluo Formula on the recovery of neurological function in MCAO rats were investigated by preparing MCAO rat model.Methods:(1)Establishment,grouping and administration of the model: 60 SD male rats were purchased and fed for one week,and the MCAO model was prepared by modified Longa thread occlusion method.According to m NSS score,rats with 5-14 scores were randomly divided into model group,sham operation group,butylphthalide group,Qixian Tongluo recipe high dose group,Qixian Tongluo recipe medium dose group and Qixian Tongluo decoction low dose group.Intervention administration began on the 3rd day after operation,and the drug was administered continuously for 14 days.(2)Index detection: the neurological function was evaluated on the 3rd,7th and 14 th day after operation by m NSS score table.At the end of the intervention,the samples were collected on the 14 th day,and the relative expressions of SYN and PSD-95 in the peri-infarcted area of MCAO rats were detected by immunohistochemistry,and the relative expression of Ephrin B2 and eph B2 proteins in the peri-infarcted area of MCAO rats were detected by;Western Blot.Results:(1)Neurological function score: The neurological function score of the sham operation group was 0 on the 3rd,7th and14 th day,indicating good neurological function.The neurologicalscores of the model group were all increased on the 3rd,7th and 14 th day,and the neurological function was impaired.Compared with sham operation group,the neurological function scores of TCM groups and butylphthalide groups were increased on day 7 and 14(P < 0.01),but were still lower than thatof model group at the same time point(P < 0.05).Compared with the third day,the neurological function scores of the Qi-Xian Tongluo prescription high dose group on the 7th and 14 th days decreased gradually(P < 0.01),which were lower thanthose of the Qi-Xian Tongluo prescription low dose group andthe butylphthalide group on the 7th and 14 th days(P < 0.01),and the decrease was most obvious in the high-dose group(P < 0.05).(2)Average optical density of SYN and PSD-95: Compared with sham operation,average optical density of SYN and PSD-95 in model group decreased(P < 0.01);Compared with the model group,the mean optical density values of SYN and PSD-95 in the butylphthalide group and Qixian Tongluo prescription dose groups were increased(P < 0.05).Compared with the butylphthalide group,the mean optical density values of SYN and PSD-95 in the Qixian Tongluo prescription high dose group were increased(P < 0.05),and the high dose group was themost significant(P < 0.05).(3)The relative expression levels of ephrin B2 and ephrin B2:Compared with sham operation group,the relative expression levels of ephrin B2 and ephrin B2 in Qi-Xian-Tongluo prescription groups,butylphthalide group and model group were decreased(P < 0.01),while those in Qi-Xian-Tongluo prescription groups and butylphthalide group were higher than those in model group(P < 0.01).Compared with butylphthalide and Qi-Xian-Tongluo prescription low-dose groups,the relative expression of Ephrin B2 in Qi-Xian-Tongluo prescription high-dose and medium-dose groups was increased(P < 0.01),and the increase was most significant in the high-dose group(P < 0.01).Compared with butylphthalide,the relative expression of Eph B2 in Qixian Tongluo prescription groups was increased(P < 0.01).The relative expression level of Eph B2 in the high dose group was significantly higher than that in the different dose groups of Qixian Tongluo prescription(P < 0.01).Conclusion:1.MCAO rats showed neurological dysfunction,decreased expression of synaptic structural remodeling signature proteins SYN and PSD-95 and decreased expression of Ephrin B2/Eph B2 signaling pathway proteins.2.Qixian Tongluo formula can promote the recovery of nerve function in MCAO rats,and the effect of high dose is the most significant.3.Qixian Tongluo formula upregulated the expression of Syn and PSD-95,the symbolic proteins of synaptic remodeling,and promoted synaptic remodeling,especially at high dose.4.Qixian Tonglu could promote synaptic remodeling by0 regulating Ephrin B2/Eph B2 signaling pathway,and high dose was the most significant... |
