Objective: To explore the effect of Yishen Jiannao Recipe on learning and memory ability of vascular dementia rats and the expression of BDNF/Trk B in hippocampal.Methods: 65 SPF male SD rats were selected.According to the random number table method,6 rats were randomly selected as the sham-operated group,The remaining 59 rats were established by a modified permanent common carotid artery ligation method to establish a vascular dementia rat model.After 4 weeks,Morris water maze selected 21 dementia model rats and randomly divided them into model group,traditional Chinese medicine group,and positive control group,7 rats in each group.The Chinese medicine group was given Yishen Jiannao decoction(12.15g/kg.d),the positive control group was given Donepezil suspension(0.45mg/kg.d),the model group and sham operation group were given equal volume of distilled water.Weighing once a week to adjust the dosage,continuous administration for 1month.Morris water maze test to evaluate the spatial learning and memory ability of rats 24 hours after the last gavage,and then sacrificed hippocampus tissues,HE staining observation The morphological changes of hippocampal CA1 area and Western Blot were used to detect the expression of BDNF and Trk B proteins in hippocampal.The statistical results of the experimental data are expressed as the meanąstandard deviation.The single-factor ANOVA analysis was used to conform to the normal distribution,the LSD method was used for pairwise comparision,and the Tamhane's method was used for uneven variance.p<0.05 indicates that the difference is statistically significant.Results:1.behavior test:In the positioning and navigation test,with the extension of training time,the escape latency of rats in each group was shortened to varying degree.On the first and second days of training,there was no significant difference in the average average escape latency of rats in each group(p>0.05).Starting on the third day of training,compared with the sham operation group,the average average escape latency of the model group rats was significantly prolonged(p<0.01),compared with the model group,although the average escape latency of the rats in the traditional Chinese medicine group and the positive control group tended to shorten,the difference was not statistically significant(p>0.05);by the 4th and 5th days,compared with the model rats The average average escape latency of rats in each group was significantly shortened(p<0.01);during the whole training process,the average average escape latency of rats in the Chinese medicine group was close to that of the positive control group,and the difference was not statistically significant(p>0.05).2.HE staining:The number of neurons in hippocampal CA1 area of the sham-operated group was abundant,regular in shape,closely arranged,and stained uniformly.The number of neurons in the hippocampal CA1 area of the Va D model group was significantly less than that in the sham-operated group,and the cell arrangement was disordered and loose,the cell vacuolation was obvious,and the phenomenon of mitosis or nuclear shrinkage was also seen.The neurons in the hippocampal CA1 area of the traditional Chinese medicine group and the positive control group still had a slight disorder of arrangement,and occasionally cell vacuoles and nuclear shrinkage,but the number and structure of neurons are improved compared with the model group.3.BDNF/Trk B expression:Compared with the rats in the sham operation group,the expression of BDNF and Trk B protein in the hippocampus of the model group was significantly reduced(p<0.01).Compared with the Va D rats in the model group,the expression levels of BDNF and Trk B protein in the hippocampus of the Chinese medicine group and the positive control group were significantly increased,and the differences were statistically significant(P<0.01).Conclusion:Yishen Jiannao Recipe can improve the learning and memory abilities of vascular dementia rats.The mechanism may be related to the up-regulation of BDNF/Trk B protein expression in hippocampal and repair nerve cell damage.
