Aim:Vascular Dementia,VD is a frequently encountered disease in the elderly,VD is second to the alzheimer's disease dementia type disease.Traditional Chinese medicine Dipsacus asper,also called as sichuan Dipsacus asper,for sichuan and radix dipsaci dipsacaceae plants(Dipsacus asperoides)dry root.Now the researches about its treatment for vascular dementia were less.So,in this research,By establishment of vascular dementia model,after giving extracts from radix dipsaci,to discuss the possible mechanism of action,it is for traditional Chinese medicine radix dipsaci further development and provide a reliable theoretical basis for clinical application.Methods:60 Normal adult male SD rats were selected out,then the animals were randomly divided into normal group,sham-operated group,model group,and low,middle and high dose group.Vascular dementia model was established by using the clip again and again recanalization bilateral common carotid artery and celiac injection of sodium nitrate tome.Rats in Low,middle and high dose group were given 15mg/kg,45mg/kg and 13 5mg/kg extracts of Dipsacus asper,and control group and model group rats were given distilled water for 28d.Then the general condition of rats was observed,and cognitive learning ability of each group rats were determined by using Morris maze.rat hippocampal CA1 area was observed by HE dyeing.Use western blot method to determine the expression of Bcl-2 protein and Bax protein in the hippocampus of rats.And SOD activity and MDA,and GSH-px levels in serum were determined though enzyme-linked immunoassay(ELASA)for each group.Results:1.Effect of extracts of Dipsacus asper on rats' general conditionDuring the experiment,the general condition of all rats are good.food intake,relieve,body hair of the animal constituting the mental state,etc.After the experiment,each animal have anatomy,macroscopic observe gastrointestinal organs such as liver and lung,kidney and spleen heart,no significant anomalies were found shows that extracts of Dipsacus asper does not lead to substantial organs of the general pathological damage;2.Effect of extracts of Dipsacus asper on learning and memory abilityEffect of extracts of Dipsacus asper on incubation period With the increase of training days,the incubation period of all rats were decreased,on the first and second day,the rats had no obvious difference to find the platform,but since the third day,the incubation period of sham group and normal group rats were shorten(P<0.05)compared with model group.And the incubation period of rats in high dose and middle dose extracts Dipsacus asper were significantly lower than the model group rats(P<0.05);Effect of extracts of Dipsacus asper on crossing platform number After withdrawal of the platform,the model group rats crossing platform number were significantly less than control group and normal group(P<0.05),while the s crossing platform number of rats in high dose Dipsacus asper group was obviously higher than that of model group(P<0.05)3.Effect of extracts of Dipsacus asper on hippocampus CA1 areas neurons morphology in vascular dementia ratsAfter HE staining of each rat's hippocampal,it was found that normal rats and control group rats hippocampal nerve cells in the CA1 area are closely packed neatly,the number is more,cells form a complete,normal structure,abundant cytoplasm,nucleolus,the boundaries of the nucleus and cytoplasm is visible.However,he number of neurons in hippocampal CA1 area in model group seriously reduce,irregular arrangement,the incomplete forms of structural abnormalities,few cytoplasm and cell nucleus and cytoplasm is blurred,triangle or irregular form.While in in high dose and middle dose Dipsacus asper group hippocampus CA1 area neurons form relatively regular,structure is relatively normal,the boundaries of the nucleus and cytoplasm is relatively clear,high dose group of cell morphology were more close to the control group,then middle dose and low dose.4.Effect of extracts of Dipsacus asper on expression of Bcl-2 and Bax protein in hippocampusIn the normal group and sham group,Bcl-2 and Bax protein in the rat hippocampus expression quantity is less,but these two kinds of protein expression in the model group was obviously increased(P<0.05),and high dose group of rats in the amount of the Bcl-2 protein expression in the hippocampus is higher than the model and quantity of Bax protein expression is lower than the model group with statistical significance(P<0.05);In Low dose group two kinds of protein expression in the hippocampus and no difference with the model group.In the normal group and sham group,Bcl-2 and Bax protein expression in hippocampal are almost the same,the ratio of model group significantly decreased,Bax protein expression in hippocampus and increased significantly more than the Bcl-2 protein,and in high dose group of,the amount of the Bcl-2 protein expression in the hippocampus are more than Bax expression.5.Effect of extracts of Dipsacus asper on SOD activity and MDA,and GSH-px levels in serumThe SOD activity and GSH-px levels in blood of rats in Middle and high dose group are significantly higher than the model group(P<0.05),at the same time,MDA in blood of rats is significantly lower than model group(P<0.05)and no obvious difference was found between the normal group compared with shame group(P<0.05)Conclusions:1.Extracts from Dipsacus asper could significantly improve cognitive learning and memory impairment in vascular dementia rats;2.Extracts from Dipsacus asper may play a role in protecting hippocampus neurons of vascular dementia rats by adjusting expression of Bcl-2 and Bax protein and improving the ability of free radical clearance the process of metabolism.
