ObjectiveVD model SD rats prepared by four-vessel occlusion method(4-VO method)were used as the experimental subjects.The method was used as an intervention to observe the effect of the VD model on the VD model rats learning and memory ability and PI3K/Akt signal transduction pathway,This article mainly evaluate whether electric acupuncture has an effect on the VD model rats and its ralated mechanism,to provide a new opinion on the study of VD.MethodsThirty-six SPF grade SD rats were divided into sham operation group,vascular dementia model group(model group),Tongdutiaoshen acupuncture group(electroacupuncture group),LY294002 group(LY model group),LY294002+electroacupunture(LY electroacupuncture group),DMSO+electroacupuncture group(DMSO electrocacupuncture group),Each contains 6 rats.The VD model made by 4-VO method.After the VD model was prepared,the PI3K/AKt signal transduction pathway inhibitor LY294002 was injected into the lateral ventricle of the LY model group and the LY electroacupuncture group and the DMSO electroacupunture group was injected DMSO.The rats in the control group were treated 25 minutes per day with the points of "Baihui","Dazhui" and "Spleen","shenshu".model group,LY model group without acupuncture,sham operation only for surgical trauma treatment.Aafter two weeks of treatment,Morris water maze test,divided into positioning navigation experiment and space exploration experiment.It tooks a week.The learning and memory abilities of the rats were observed.The expression of PI3K and AKt protein in the hippocampus of rats were observed by Western Blot method.The experimental data were analyzed by spss17.0.Results1.Morris water maze resultsThe escape latency of each group was different(P<0.01).Compared with the other five groups,the escape latency was significantly shortened in the sham operation group(P<0.05).Compared with the model group,the escape latency of the model group was significantly longer than that of the electroacupuncture group(P<0.05).There was no significant difference between the LY model group and the LY group(P>0.05).Compared with LY electroacupuncture group,the latency of esophageal group was significantly shortened(P<0.01).The sham was different when it comes to the frequency of passing through the original platform compared with the LY model group and the LY electroacupunture group(P<0.05),As the same when these two LY groups compared with the electroacupuncture group.But there were no differents when the model group compared with these two LY groups.And The frequency of passing through the original platform of the rats in the DMSO electroacupuncture group was significantly higher than that in the LY group(P<0.01).2.Western Blot resultsWestern Blot showed that the protein contents of AKt and PI3K increased in the sham operation group compared with the model group,LY electroacupuncture group and LY model group(P<0.01),but compared with the electroacupuncture group and the DMSO electroacupuncture group,there were no significance(P>0.05).The content of PI3K and Akt protein in hippocampus of electroacupuncture group significantly higher than that of model group(P<0.01),There was no significant difference between the LY model group and the LY group(P>0.05),but both of these two groups were lower than the model group.ConclusionsElectroacupunture at "Baihui","Dazhui" and "Spleen","shenshu" can improve the learning and memory ability of vascular dementia model rats and can promote the recovery of learning and memory ability in vascular dementia model rats.electroacupunture at "Baihui","Dazhui" and "Spleen","shenshu"can increase the PI3K/Akt signal transduction pathway in PI3K?Akt protein expression,Thus promote cell proliferation,differentiation,inhibition of this pathway-related pro-apoptotic protein expression,reduce ischemia on hippocampus injury in rats,play a protective role of nerve cells,which may be Tun One of the mechanisms of treatment of vascular dementia.
