Purpose:To establish chronic heart failure rats models(CHF)and curing Yi Qi Huo Xue compound under the imfluence of TGF-β1,to explore the effective mechanism and target of function of Yi Qi Huo Xue compounds to the myocardial fibrosis of chronic heart faiture rats,which is benefits to the prevention-treatment of chronic heart failure and provides an effective theoretical basis and reference.Material and Method:1.To establish chronic heart failure rat model: selecting 100 healthy 3D male rats(SPF grade,12 weeks old,weighing between 230 and 260 g)and establish chronics heart failutr rats models by ligating anterior descending branch of coronary artery and collaboration with exhausted swim,less food and other methods.The results of electrocardiogram and echocardiography examination showed ST-T elevation,left ventricular ejection fraction(EF)≤ 45%(EF)or CI ≤ 180ml/min/kg is the measure of chronic heart failure rats moels was successful or not.2.Grouping of operation animals:selecting 10 rats randomly and taking them as sham operation group(normal group)by random numbering.Rats with successful modeling were divided into model group,Chinese medicine group and western medicine group to ensure the samples of each groupis balance with 10 rats..3.Experimental drugs: The medicines include Chinese herbal medicines Yi Qi Huo Xue compound(Huangqi,Honghua,Panax ginseng,Motherwort,Salvia miltiorrhiza,Chinese wolfberry,Chinese wolfberry).All medicines are from the Center of Traditional Chinese Medicine Preparations of the Affiliated Hospital of Liaoning University of Traditional Chinese Medicine.The production company of lisinopril tablets is Shanghai Xinyi Vientiane Pharmaceutical Co.,Ltd.,with a specification of 10 mg per tablet and the national drug license HI0960265.4.Experimental dosage of administration: In this experiment,the sham operation group or model group were not treated with any drugs but dealing with equal volume(3ml)of distilled water for intragastric administration once a day;treatment groups(Chinese medicine group,western medicine group)fed with Yi Qi Huo Xue compound 9.2g medicines / Kg / d and rats in western group fed 1.5mg lisinopril / Kg / d for 28 days in accordance with the equivalent dose of human and rats conversion.5.Test detection technology: The method used to measure the mRNA and protein expression changes of TGF-β1 in myocardial tissue of each group of chronic heart failure rats is protein immunofluorescence(Western blot),quantitative fluorescence PCR technology,and records the mean gray value;the method of measuring the concentration of BNP is through ELISA;Microscopic observation of myocardial cell morphology changes.Results:1.Compared with the other three groups,concentration of BNP in the sham operation group was the lowest,p<0.05,and the datas amoung groups were significantly different,which was proved that the level of BNP in plasma increased when heart failure occurred.Compared with the model group,the BNP concentration in treatment group decreased,p <0.01,and the datas amoung groups were significantly different,which proved that drug intervention can reduce the production of BNP in myocardial tissue.Compared with the western medicine group,the plasma BNP content was comparable,P>0.05,and the datas were different amoung groups..2.Compared with the other three groups,the sham-operated group had the lowest TGF-β1 mRNA level,P<0.01,and there was a significant difference between the two groups,indicating that the expression of TGF-β1 mRNA in the myocardial tissue was increased when the heart failure occurred.Compared with the traditional Chinese medicine group and the Western medicine group,the TGF-β1 mRNA content in the model group was the highest,P<0.05,there was a difference between groups,it was proved that Chinese and Western drugs can interfere with the expression of TGF-β1 mRNA,and all of them have chronic heart failure.Significant effect;Chinese medicine group compared with the Western medicine group,TGF-β1mRNA content is quite,P> 0.05,the difference between the groups was not statistically significant,prove that Chinese and Western medicine treatment of chronic heart failure are effective,and the efficacy is equivalent.3.Compared with the other three groups,the TGF-β1 protein level of sham group was the lowest,P<0.01,and there were significant differences amoung groups,which indicated that the expression of TGF-β1 protein in myocardial tissue increased with chronic heart failure.The degree of myocardial fibrosis was aggravated.Compared with the treatment group,the TGF-β1 protein content in the model group was the highest,P<0.05,there were differences amoung groups,which indicated that both Chinese and Western medicines can interfere with the expression of TGF-β1 protein.Chronic heart failure has a significant effect;compared with the Western medicine group,the TGF-β1 protein content is comparable,P>0.05,which can be negligible.The above comparisons can beconcluded that Chinese and Western medicines have effects on the treatment of chronic heart failure and the difference between Chinese and western medicines were slight.4.Changes in morphology of myocardial cells under microscope: In the sham-operated group,the myocardial cells in the sham-operated rats were arranged closely and neatly,the shape and size of the nucleus were comparable,the transverse stripes were clear,the myofibrils were distributed in an orderly manner,and there were no fractures and defects;the myocardial cells in the model group were intricate and complicated.The size of the shape was irregular,the transverse stripes disappeared,and the myocardial fibers were disorderly and disordered.The rats in the treatment group exhibited regular arrangement of myocardial cells,similar nucleus size,orderly arrangement of myofibrils,visible transverse stripes,and fractures or defects.Thus,Yi Qi Huo Xue compound can inhibit myocardial fibrosis and prevent ventricular remodeling,and it has curative effect in the treatment of chronic heart failure,and the efficacy is comparable..Conclusion:1.Compared with the concentrations of BNP in each group,the results demonstrate that BNP concentrations increased when chronic heart failure occurred.2.In the model group,the protein and mRNA expression levels of TGF-β1 were higher than the rest of the groups,which proved that TGF-β1 is highly expressed during the onset of chronic heart failure,participated in myocardial fibrosis and promote the occurrence and development of chronic heart failure.3.Compared with the model group,the protein and mRNA expression levels of TGF-β1 in the Chinese medicine group and the Western medicine group were lower,which indicated that the Chinese and Western medicines can inhibit myocardial fibrosis and delay the occurrence and development of chronic heart failure.4.There was no significant difference in protein and mRNA expression levels of TGF-β1 between Chinese medicine group and western medicine group,which was proved that both Chinese and Western medicines can interfere with myocardial fibrosis,and the curative effect of chronic heart failure is equivalent,and it is worth popularizing and applying.5.Through the comparison between the groups,the morphology of the myocardium under the microscope was changed,which proved that the form of the myocardial cells was destroyed when heart failure occurred.
