| Purpose:In this study,we established a rat model of chronic heart failure after myocardial infarction to observe the effect of Yiqi Huoxue Recipe on the expression of mitochondrial transcription factor A m RNA(mt TFA m RNA)and mitochondrial transcription factor A protein(mt TFA protein)in rats with chronic heart failure.Western blot,real-time fluorescent quantitative PCR(RT-PCR)and other experimental methods,from the perspective of genes,proteins and histomorphology,explore the mechanism of action of Yiqi Huoxue Recipe on chronic heart failure.Medicine provides a reliable and effective evidence for the prevention and treatment of chronic heart failure.Material and method:1.Model of chronic heart failure in rats by ligation of coronary artery combined with food reduction,exhaustive swimming,etc.,observe the activity of rats,by means of ultrasound Doppler measurement: left ventricular ejection fraction(EF)≤ 50% And the heart index(CI)≤ 180ml(min · kg)to determine the success of chronic heart failure modeling.2.The heart failure rats with successful modeling were divided into three groups: model group,western medicine group and Chinese medicine group.Normal rats that were not surgically modeled were set as a blank control group.There were 10 rats in each group in the model group,western medicine group,Chinese medicine group and blank control group.The rats in the model group and the rats in the blank control group were not treated with drugs.The normal saline volume of the rats in the two groups was 6 ml/day.Each group of rats can be administered according to the conversion formula of the equivalent dose of human and mouse,and the dose(g/Kg)= human dose(g)× 0.018 / animal body weight(Kg).The Chinese medicine group was 9.2 g(raw drug)/ Kg / d;the western medicine group was(lisinopril)1.5 mg / Kg / d,twice a day,continuous administration for 4 weeks.3.Real-time quantitative PCR(RT-PCR),Western blot(Western blot),electron microscopy and enzyme-linked immunosorbent assay(ELISA)were used to compare chronic heart failure rats(model group,blank control group,The expression of mitochondrialtranscription factor A m RNA(mt TFA m RNA)and mitochondrial transcription factor A protein(mt TFA protein)in the myocardium and the morphological changes of related myocardial tissue were observed in the Western medicine group and the Chinese medicine group.Results:1.The mitochondrial transcription factor A m RNA was significantly decreased in the animal model group with chronic heart failure,compared with the blank group(P<0.05).The expression of mitochondrial transcription factor A m RNA was significantly increased in the western medicine group and Yiqihuoxue compound group,compared with the model group P<0.05.Compared with the western medicine group,the Yiqihuoxue compound group had P>0.05,which was not statistically significant.2.The expression of mitochondrial transcription factor A protein in the chronic heart failure animal model group was significantly decreased compared with the blank group(P<0.05).The expression of mitochondrial transcription factor A protein in the western medicine group and Yiqihuoxue compound group was significantly higher than that in the model group.0.05;Yiqihuoxue compound group compared with western medicine group P>0.05,no statistical significance.3.The expression of BNP in the chronic heart failure animal model group was significantly higher than that in the blank group(P<0.05).The expression of BNP in the Western medicine group and Yiqihuoxue compound group decreased significantly,compared with the model group P<0.05;Yiqihuoxue compound group Compared with the western medicine group,P>0.05,no statistical significance.4.Under the electron microscope,the cardiomyocytes of the blank control group were arranged neatly,the interstitial edema was absent,the membrane and nucleus were clear,the boundary between the light and dark bands was clear,the number of mitochondria was abundant,the structure of the mitochondria was clear,the matrix was closely arranged,and the shape of the inner ridge was clearly visible.The original granules were intact;the myocardial fibers in the model control group were disordered,the muscle fibers were broken and disappeared,the muscle fiber interstitial and the nucleus were obviously edematous,themitochondria were reduced and swollen,the internal ridge structure was disordered or disappeared,and the amount of glycogen was reduced.The cell nucleus was swollen and the sarcoplasmic reticulum was dilated.The myocardial fiber structure of the traditional Chinese medicine group and the western medicine group was improved compared with the model group.The myocardial tissue edema was reduced to some extent,the amount of glycogen was increased,and mitochondrial vacuolization and mitochondrial dysfunction were observed locally.The range of myofilament dissolution and necrosis is reduced,and the structure of mitochondria is also improved to varying degrees.It is indicated that Yiqi Huoxue Recipe can promote the recovery of mitochondria in both quality and quantity,and the mitochondrial structure abnormality is improved,and the efficacy of Yiqi Huoxue Recipe group is similar to that of western medicine group.Conclusion:1.Rats were subjected to coronary artery ligation combined with exhaustive swimming and starvation to reduce food.The cardiac function of rats was detected by echocardiography,which proved that the method can be used for modeling rats with chronic heart failure.2.Rats with chronic heart failure were treated with Yiqi Huoxue Recipe for 4 weeks.Western blot,real-time quantitative PCR(RT-PCR)and electron microscopy were used to detect myocardial tissue.The results showed that: Model group Yiqi Huoxue Recipe can promote the expression of mitochondrial transcription factor A m RNA and mitochondrial transcription factor A protein in myocardial tissue of rats with chronic heart failure.Yiqi Huoxue Recipe can regulate mitochondrial metabolism,thereby improving myocardial energy metabolism,so that ventricular remodeling is inhibited and heart function is restored.3.The above experimental results are sufficient to show that Yiqi Huoxue Recipe is safe and reliable in the treatment of chronic heart failure,and its therapeutic effect is similar to that of western medicine lisinopril. |
