Study On The Quality Standard Of Periplaneta Americana Medicinal Materials

Objective:As a bulk drug for several listed drugs,Periplaneta Americana has a very high medicinal value;however,the Periplaneta Americana is not included in the Chinese Pharmacopoeia,and the current local standards have certain differences.Therefore,this paper systematically studies the quality of medicinal materials in the Periplaneta Americana.Based on the Chinese Pharmacopoeia and local standards,the research uses modern scientific research techniques to analyze and study the identification,inspection and content determination of medicinal materials,supplement and improve the specific quality standard methods and limits,and to improve and supplement the research.Local standards,the development of a more complete and reliable quality standards for Periplaneta Americana,provides a basis for the entry of Periplaneta americana into Chinese Pharmacopoeia standards.Method:1.Microscopic identification was used to find the microscopic characteristics of the Periplaneta Americana medicinal materials.The local conditions were used to identify the authenticity of the thin-layer color sputum.2.In addition to the local standards of the Periplaneta Americana and the Chinese Pharmacopoeia inspection method,the impurities,moisture,ash,leachate,heavy metals,sulfur dioxide,pesticide residues and aflatoxins under the inspection of the Periplaneta Americana are verified,supplemented and improved.3.To control the multi-component content of Periplaneta Americana,the content of total polysaccharides and water-soluble proteins was determined by Ultraviolet Spectrophotometry after ultrasonic extraction.The content of eight amino acids in Periplaneta Americana was determined by pre-column derivatization with high performance liquid chromatography;and methodological investigation was carried out for the determination of 20 common amino acids in the alcohol extract of Peplaneta Americanria;automatic amino acid analyzer was used to determine 20 common amino acids in the alcohol extract of Periplaneta Americana.The content of nucleotides was determined by high performance liquid chromatography with uracil,hypoxanthine,xanthine and inosine as control.he method for the determination of the content of nucleotides was discussed.Methods Methodological investigation was carried out.Result:1.Identification:The microscopic identification of the Periplaneta Americana is obvious,and the foot fragments,bristles,body fragments and wing fragments can be clearly observed.Using n-butanol-glacial acetic acid-water-methanol（19:5:5:3）as developing agent and 0.5%ninhydrin at 105°C for co-heating,15 batches of Periplaneta Americana were obtained.There are 6 common spots in the same position.15 batches of Periplaneta Americana are authentic.2.Inspection:The impurity content of Periplaneta Americana was 0.1088%⁰.346%,the average content was 0.243%;the moisture content was 4.447%¹0.429%,the average content was 6.973%;the ash content was 3.507%⁶.624%,the average content was 5.605%;the acid insoluble ash content was 0.333%⁰.597%,the average content was 0.460%;the extract content was 8.548%¹3.163%,the average content was 10.398%.The contents of copper,arsenic,cadmium,lead and mercury were9.054⁶6.448 mg/kg,the average content was 25.553 mg/kg,0.190⁰.630 mg/kg,0.517 mg/kg,the average content was 0.009⁰.041 mg/kg,0.019 mg/kg,0.096¹.229 mg/kg,the average content was 0.734 mg/kg,0⁰.394 mg/kg and the average content was 0.086 mg/kg,respectively.The residues of SO₂ in the Periplaneta Americana were 35 mg/kg to 87 mg/kg,with an average content of 56.53 mg/kg.Dichlorvos,alpha-HCH,pentachloronitrobenzene,gamma-HCH,beta-HCH,dimethoate,delta-HCH,p,p’-DDE,o,p’-DDT,p,p’-DDD,p,p’-DDT,cypermethrin were not detected in Periplaneta Americana.Aflatoxin B2,aflatoxin G1 and aflatoxin G2 were not detected in Periplaneta Americana.Aflatoxin B1 was lower than the detection limit of 0.150μg/kg.3.Content determination:The total polysaccharide content in Periplaneta Americana was 4.21 mg/g⁷.37 mg/g,with an average content of 5.52 mg/g;the water soluble protein content was 21.08 mg/g³5.96 mg/g,with an average content of 28.20 mg/kg;after PITC derivation,the total content of glutamic acid,glycine,histidine,alanine,valine,isoleucine,lysine in 15 batches of Periplaneta Americana was 1.44%.The average content of Uracil,Hypoxanthine and Xanthine in 15 batches of Periplaneta Americana was 0.22%¹.81%,and the average content was 0.70%.The total content of 20 free amino acids in 15 batches of Periplaneta Americana was 10.00%¹6.02%,and the average content was 12.37%.Conclusion:In this paper,a more comprehensive quality-related research was carried out on the Periplaneta Americana,and the experimental research was carried out from the aspects of identification,inspection and content determination.The standards of the Periplaneta Americana contained in the local standards were compared,and the pharmacology of the Periplaneta Americana was increased.The material quality control indicators of the drug efficacy were controlled and the thin layer development conditions were optimized for the thin layer identification items,which verified the superiority of local standards to its quality control.The control limits for moisture and total ash content in the inspection items were improved,and heavy metals,sulfur dioxide,pesticide residues and aflatoxins were examined.Through this study,it is proposed to establish a more practical standard that can respond to better quality of the Periplaneta Americana.
