Objective: To observe the influence of aerobic exercise on memory ability and the expression of p CREB,PICK1 and Glu R2 on hippocampus in rats with ischemic cognitive disorder.and exploring the interaction mechanisms of which.Methods: 56 male Sprague-Dawley rats were randomly divided into four groups sham-operation group:model group,medicinal group and aerobics group according to the numeration table(n=14 in each group).Then model group,medicinal group and aerobics group all established the model by permanent ligature of bilateral common carotid artery.All groups were evaluated by Morris water maze,and Morris water maze was used to evaluate escape latency and number of crossing platforms.Record the times of escape latency and the number of crossing platforms for each Morris water maze.The pathomorphological changes of CA1 areas in hippocampus was observed by Nissl staining and the impression of p CREB,PICK1 and Glu R2 was also measured by western blot.Results:1.General condition: Rats are active,circadian rhythm and has normal diet before modeling.After modeling,the rats showed decreased excitement,sensitivity to various stimuli,less movement and supine position,sluggish eyes and slow weight gain,which improved after treatment.The rats were weighed weekly from the day before modeling.In sham operation group,the body weight of rats showed a steady upward trend,while the remaining three groups on the second day after modeling weight loss but gradually rise.Then there was no significant difference in body weight at the same time in each group(P>0.05).2.Morris water maze assessment results show that:(1)In sham operation group,swimming routes tend to be more linear and more targeted;The swimming route of model rats which concentrated in surrounding areas was obviously random and circuitous fold.It belongs to the edge type swimming search strategy;The acupoint catgut embedding group and aerobic exercise group rats had swimming routes which was also close to the edge region slightly but shorter than the model group.(2)The average escape latency was longer and the platform search frequencies decreased significantly in model group than sham group(P<0.01);The time rats need to find platform was significantly shorter(P<0.01)and the platform search frequencies was more(P<0.05)in aerobic exercise group and catgut embedding group than model group.There is no significant difference between the aerobic exercise group and catgut embedding group(P>0.05).3.To observe the morphology of hippocampus tissue CA1: in spite of sham operation group,the other three groups has fewer Nissl bodies whose structure is not clear and mess and damaged in varying degrees;The structure of cells was damaged partly and the number of nissl bodies was more in aerobic exercise group and catgut embedding group than in model group.There is little difference between the two groups.4.Western blot showed that: the p CREB and Glu R2 protein expression in the hippocampus of rats decreased while PICK1 protein expression increased significantly in model group than sham operation group(P<0.01);the p CREB and Glu R2 protein expression increased(P<0.05,P<0.01)while the PICK1 protein expression reduced(P<0.01)in the aerobic exercise group and catgut embedding group than model group.There is no significant difference between the aerobic exercise group and catgut embedding group(P>0.05).Conclusion:1.The aerobic exercise can effectively enhance the learning and memory ability of rats with chronic ischemic cognitive disorder.2.Part of the mechanism of aerobic exercise therapy to improve cognitive function may be to promote phosphorylation of CREB,thereby down regulating the expression of PICK1 in hippocampus and up regulating the content of subunit Glu R2,and ultimately achieve the effect of promoting synaptic plasticity and improving cognitive function such as learning and memory.
