Objective: The insomnia rats were used as the subjects,using a fixed local acupoints、acupoints with different meridians for comparative study,To observe the effects of acupuncture treatment at different meridians on the levels of HPA axis related hormones、5-HT1a and 5-HT2a receptor expression in hypothalamus and acupoint compatibility in insomniarats;To discuss the mechanism of action for acupuncture treatment.Methods: Chlorophenylalanine suspension was under intraperitoneal injection to establish insomnia model rats.60 SD rats were randomly divided into 5 groups,including blank group,model group,Baihui + Shenmen group,Baihui + Sanyinjiao group,Baihui + non-acupoint group,with 12 rats in each group.Each treatment group received acupuncture in relevant acupoints,30 min each time,for 7 d.ELISA was used to measure the levels of CRH,ACTH and CORT;real-time fluorescence quantitative PCR was used to measure The expression of 5-HT1a and 5-HT2a mRNA in hypothalamus.Results: 1.Before and after modeling,pentobarbital sodium positive test: There was no significant difference in sleep latency and total sleep time before and after Blank group modeling(P> 0.05).After injection of sodium pentobarbital in model group,the latent period of sleep after modeling was significantly more than that before modeling,the difference was statistically significant(P <0.05).The total sleep time was significantly less than before modeling,the difference was statistically significant(P <0.05),suggesting that modeling success.2.Before and after treatment,pentobarbital sodium positive test: Sleep latency comparison: Compared with the blank group,the model group was delayed(P <0.05).Compared with the model group,the three treatment groups were advanced(P <0.05).There was no significant difference between the treatment groups(P> 0.05).Sleep time comparison: Compared with the blank group,the model group decreased(P <0.05);Compared with model group,Baihui + Shenmen group prolonged(P <0.05),but there was no significant change in Baihui + Sanyinjiao group and Baihui + non-acupoint group(P> 0.05).Comparison between the treatment group,Baihui + Shenmen group leader in Baihui + Sanyinjiao group and Baihui + non-non-acupoint group(P <0.05).3.The effects of acupuncture treatment at different meridians on the levels of HPA axis related hormones:Compared with the blank group,the contents of CRH in the hypothalamus,ACTH and CORT in serum of model group were significantly increased(P <0.01).Compared with model group,the levels of CRH in the hypothalamus,ACTH and CORT in serum of Baihui + Shenmen group and Baihui + Sanyinjiao group were significantly decreased(P <0.01),The levels of serum ACTH in Baihui + non-acupoints group were significantly decreased(P <0.05),while the levels of serum CORT were significantly decreased(P <0.01),and there was no statistical difference in the hypothalamus CRH in this group(P> 0.05).Compared with Baihui + Shenmen group,there was no significant difference in hypothalamus CRH content in Baihui + Sanyinjiao group and Baihui + non-acupoint group(P <0.05),serum ACTH content was significantly increased(P <0.01),and serum CORT content was significantly increased(P <0.05).Compared with the Baihui + Sanyinjiao group,the content of CRH in the hypothalamus of Baihui + non-acupoint group was significantly increased(P <0.05),the serum CORT content was significantly increased(P <0.01),but the serum ACTH content was not statistically different Significance(P> 0.05).4.The effects of acupuncture treatment at different meridians on the expression of 5-HT1a and 5-HT2a receptor in hypothalamus: Compared with the blank group,the expression of 5-HT1a receptor mRNA in the hypothalamus of model group was significantly decreased(P <0.01)and the expression of 5-HT2a receptor mRNA was significantly increased(P <0.01),with statistical significance;Compared with the model group,the expression of 5-HT1a receptor mRNA in Baihui + Shenmen group was significantly increased(P <0.01),while the expression of 5-HT2a receptor mRNA was significantly decreased(P <0.01).Compared with Baihui + Shenmen group,the expressions of 5-HT1a receptor mRNA in Baihui + Sanyinjiao group and Baihui + non-acupoint group were significantly decreased,(P <0.05)and(P <0.01),respectively,with statistical significance.The expression of 5-HT2a receptor mRNA in Baihui + Sanyinjiao group had no significant change(P> 0.05),and the expression of 5-HT2a receptor mRNA in Baihui + non-acupoint was significantly increased,has statistical significane;Compared with Baihui + Sanyinjiao group,there was no significant difference in the expression of 5-HT1a and 5-HT2a receptor in Baihui and non-acupoints group.Conclusion:Acupuncture may play a role in sedative and soothing by regulating the hormone levels related to HPA axis and the mRNA expression of 5-HT1a and 5-HT2a receptors.acupuncture acupoints at different meridians may be one of the factors to determine the difference of acupoints synergy effect. |
