Objective: To compare the acupoints of acupoints with different meridians and acupoints by fixing acupoints,and to observe the effect of acupuncture on the contents of monoamine neurotransmitters 5-hydroxy tryptamine,Dopamine and Norepinephrine and the expressions of ventrolateral preoptic MT1 and MT2 mRNA in insomnia rats,and discussion effect of acupuncture on insomnia and the effect of acupuncture on the compatibility of acupointsMethods : 60 SD rats were randomly divided into normal control,model,same meridian:“Bahui”(GV 20)-“Shenmen”(HT 07,3-acupoints),related meridian : “Bahui”(GV 20)-“Sanyinjiao”(SP 06,3-acupoints),non-acupoint :“Bahui”(GV 20)-non-acupoint(3-acupoints)groups,with 12 rats in each group.Insomnia model was eatablished by Intraperitoneal injection of para chlorophenylala-nine suspension.Acupuncture therapy once daily for 7 days,the contents of NE,DA and 5-HT in serum and MT1 and MT2 mRNA expression in VLPO were detected by Elisa and RT-PCR respectively.Results : 1 、 Before and after the establishment of pentobarbital sodium,there was no statistically significant difference in sleep latency and total sleep time between the blank group and the model group(P>0.05).After the injection of sodium pentobarbital in the model group rats,Compared with before,the difference in sleep latency and total sleep time after modeling was statistically significant(P<0.05).The above indicates successful modeling.2、Before and after treatment,pentobarbital sodium positive test: Sleep latency comparison: Compared with the blank group,the model group was delayed(P <0.05).Compared with the model group,the three treatment groups were advanced(P <0.05).There was no significant difference between the treatment groups(P> 0.05).Sleep time comparison: Compared with the blank group,the model group decreased(P <0.05);Compared with model group,Baihui + Shenmen group prolonged(P <0.05),but there was no significant change in Baihui + Sanyinjiao group and Baihui + non-acupoint group(P> 0.05).Comparison between the treatment groups,Baihui + Shenmen group leader in the Baihui + Sanyinjiao group,Baihui + non-acupoint group(P <0.05);between Baihui + Sanyinjiao group,Baihui + non-acupoint group Significant differences.3、 Effects of acupuncture at acupoints on selection of monoamine neurotran-smitters and expression of melatonin MT1 mRNA and MT2 mRNA in insomnia rats:Compared with the blank group,the contents of NE,DA and 5-HT in the model group increased significantly(P<0.01),and the expression of MT1 and MT2 mRNA in the VLPO group decreased significantly(P<0.01).Compared with the model group,the contents of NE and DA in the three treatment groups were significantly lower(P<0.05,P<0.01);the contents of 5-HT in Baihui and Sanyinjiao groups were significantly lower(P<0.05),and the other two groups were 5-Although there was a decreasing trend in HT content,there was no statistical significance(P>0.05).The mRNA expression of MT1 and MT2 in the VLPO of the three treatment groups were significantly higher(P<0.01).Comparison between the treatment groups:Compared with Baihui + Shenmen group,the changes of NE content in Baihui + Sanyinjiao group and Baihui + non-acupoint group were statistically significant(P <0.05);DAI content in Baihui + Sanyinjiao group had no significant difference(P> 0.05),but there were significant differences between the two groups(P <0.01)in the Baihui group and the non-acupoint group <0.01)(P> 0.05).The expressions of MT1 mRNA and MT2 mRNA in Baihui + Sanyinjiao group and Baihui + non-acupoint group were all statistically significant(P <0.05).Compared with Baihui + Sanyinjiao Group,the content of NE,DA and 5-HT and the expression of MT1 mRNA and MT2 mRNA in Baihui + non-acupoint group were significantly different(P <0.01 or P <0.05).The above indicates that Baihui + Shenmen group is superior to Baihui + Sanyinjiao group and Baihui + non-acupoint group in regulating monoamine neurotransmitters and MT1 mRNA and MT2 mRNA expression of insomnia rats.Conclusion:Acupuncture to improve insomnia in rats with serum monoamine neurotransmitter content and hypothalamic preoptic area melatonin MT1 mRNA,MT2 mRNA expression,acupoints are the factors that affect the acupoint compatibility,according to the acupoints can improve Acupoint compatibility effect.
