Objective: This study analyzed the clinicopathological features of the disease by reviewing the clinicopathological data of 88 patients with endometrial carcinoma(EC)diagnosed by pathology in the Affiliated Hospital of Qing Hai University.By detecting the expression of IMP2,PTEN,P53 and P16 in EC,it analyzes its correlation with clinicopathological features of EC as well as study the relevance of their expression in EC.At last,It explores the auxiliary diagnostic value of IMP2 expression in distinguishing endometrioid and serous carcinomas.Methods: By research on the paraffin embedded specimens of 88 patients with EC diagnosed by pathology in the Affiliated Hospital of Qing Hai University from January 2015 to September 2019 to retrospective analyze the indexes in which all patients had complete clinicopathological data.Immunohistochemical(IHC)method was used to detect the expression of IMP2,PTEN,P53 and P16 in the 88 cases EC,and recorded the data.SPSS 25.0 software was used for statistical analysis of the data..Results:1.In this study,the age of onset of patients was 35-86 years old,the average age of onset was 56.9 ±9.6 years old,the median age of onset was 56.5 years old.There were 72 patients over 50 years old(81.82%),69 patients with endometrioid carcinoma(79.31%),56 patients with depth of myometrial invasion of less than 1/2(63.64%),8 patients with lymph node metastasis(9.09%),72 patients with pathological stages Ⅰ-Ⅱ(81.82%).2.There was statistically significant difference in the expression of IMP2 in EC between patients of different ages(P<0.05),the positive degree of IMP2 expression increased with age from<50 years old group to≥50 years old group(Gamma=0.607,P < 0.05).There was statistically significant difference in the expression of IMP2 between endometrioid carcinoma and serous carcinoma(P<0.001),and the expression of IMP2 in all serous carcinoma was strongly positive.In addition,the expression of IMP2 in histological grade group was statistically significantly different,(P<0.001),and the positive degree of IMP2 expression increased with the increase of histological grade(Gamma=0.595,P<0.001).The expression of IMP2 in EC between different lymph node metastasis,depth of myometrial invasion and FIGO stage was no statistically significant difference(P>0.05).3.There was statistically significant difference in the expression of P16 in EC between different pathological types(P<0.001).There was statistically significant difference in the expression of P16 between different histological grading(P<0.001),and the positive degree of P16 expression increased with the increase of histological grade(Gamma=0.602,P<0.001).There was statistically significant difference in the expression of P16 in EC between different depth of myometrial invasion(P<0.05),and the positive degree of P16 expression increased with the depth of myometrial invasion from<1/2 group to≥1/2 group(Gamma=0.397,P<0.05).The expression of P16 in EC between different ages,lymph node metastasis and FIGO stage was no statistically significant difference(P>0.05).4.There was statistically significant difference in the expression of P53 in EC between different pathological types(P<0.001),and the proportion of the abnormal staining pattern of P53 in serous carcinoma(83.3%)was significantly higher than that in endometrioid carcinoma(17.40%).The expression of P53 between different histological grading was statistically significant difference(P<0.05),and the proportion of the abnormal staining pattern of P53 in all staining pattern increased with the increase of histological grade.There was no statistically significant difference in the expression of P53 in EC between different ages,depth of myometrial invasion,lymph node metastasis and FIGO stage(P>0.05).5.There was statistically significant difference in the expression of PTEN in EC between different pathological types(P < 0.05).There was statistically significant difference in the expression of PTEN between different histological grading(P<0.05),and the positive degree of PTEN expression increased with the increase of histological grade(Gamma=0.337,P<0.05).The expression of PTEN in EC between different ages,depth of myometrial invasion,lymph node metastasis and FIGO stage was no statistically significant difference(P>0.05).6.The expression of IMP2 was positively correlated with the expression of P16 and PTEN in EC,and the correlation coefficient was 0.400 and 0.237 respectively(P<0.05).The expression of P16 and PTEN in EC was positively correlated with the correlation coefficient of 0.321(P<0.05).7.The result of differential diagnoses of endometrioid carcinoma and serous carcinoma by IMP2 was consistent with that of original pathological diagnosis.(P>0.05).After the Kappa consistency analysis,it can be considered that the diagnostic results of IMP2 and the original pathological diagnosis are highly consistent in the differential diagnosis of endometrioid carcinoma and serous carcinoma(Kappa=0.993,P<0.001).The diagnostic rate of IMP2 detection method for endometrioid carcinoma was 97.10%,the misdiagnosis rate was 0,and the missed diagnosis rate was 2.90%.The diagnostic rate of serous cancer was 100.00%,the misdiagnosis rate was 11.11%,and the missed diagnosis rate was 0.Conclusion:1.The expression patterns of IMP2,P16,P53 and PTEN in EC were related to the pathological classification and histological grade of EC.The positive degree of IMP2,P16 and PTEN increased with the increase of histological grade,and the proportion of the abnormal staining pattern of P53 in all staining pattern of P53 increased with the increase of histological grade.It is suggested that they may be involved in the progression of EC,which has a certain reference value for judging the malignant degree of EC.2.There is a positive correlation between the expression of IMP2,P16 and PTEN in EC,suggesting that they may have a correlation in the progression of EC.3.IMP2 may be a new auxiliary diagnostic marker in pathological differential diagnosis of endometrioid carcinoma and serous carcinoma,and has auxiliary diagnostic value in distinguishing these two types of EC.
