Efficacy Of Celecoxib Adjunctive Therapy With Fluoxetine On Depression

Objective: Using Hamilton Depression Scale and plasma interleukin-6 as indicators to observe the difference of efficacy between fluoxetine combined with celecoxib and fluoxetine combined with placebo in the treatment of depressionMethods: According to DSM-IV diagnostic criteria for depression,121 patients with depression admitted to the First Affiliated Hospital of Xiamen University from September 2016 to December 2018 were randomly divided into two groups.59 patients in the first group was treated with fluoxetine combined with celecoxib(Celecoxib group),and 62 patients in the the second group was treated with fluoxetine combined with placebo(placebo group).The levels of plasma interleukin-6(IL-6)and Hamilton Depression Scale(HDS)scores were measured before treatment,at the end of the first week,at the end of the second week,at the end of the fourth week,at the end of the sixth week and at the end of the eighth week individually,the changes of mood,appetite,sleep,weight,quality of life and social relations were also observed.Results: There were no significant differences in age,sex,height,weight and onset age between celecoxib group and placebo group.At the end of the first week of treatment,there was no significant difference in the score of Hamilton Depression Scale between celecoxib group and placebo group(p=0.45);the level of plasma interleukin-6 in celecoxib group decreased significantly,and the level of plasma interleukin in placebo group also decreased,with a significant difference between the two groups(p<0.001);and at the end of the second week of treatment,the level of plasma interleukin of celecoxib group was significantly lower than that in placebo group(p<0.001).Milton Depression Scale score was significantly lower than that of placebo group(p=0.037).At the end of 4th and 6th weeks of treatment,the score of Celecoxib group was significantly lower than that of placebo group(p<0.001).At the end of 2nd,4th and 6th weeks of treatment,the level of plasma interleukin-6 in Celecoxib group was significantly lower than that before treatment(p<0.001).There was a significant decrease in plasma interleukin-6 in placebo group,and there was a significant difference between the two groups(p < 0.001).Conclusion: Celecoxib may play an anti-inflammatory role by reducing the level of plasma interleukin-6 in patients with depression and than enhance the efficacy of fluoxetine on depression.
