Background:Alzheimer's disease(AD)is a common neurodegenerative disease characterized by cognitive decline.It has gradually become one of the world's leading disabling and lethal diseases.However,there is currently no effective treatment method except early clinical intervention,so early diagnosis method is important.The extracellular amyloid deposit formed in the cerebral cortex of AD patients is its unique pathological changes of AD patients,and ?-amyloid 42(A?42)as its main component has attracted much attention in the application as diagnostic biomarker.According to AD clinical guidelines,the concentration of cerebrospinal fluid A?42 has been used as one of the indicators during AD diagnosis.However,due to the potential risks of lumbar puncture,the development of blood diagnostic method is urgently needed in clinical.Multiple studies of blood levels of A?42 in AD patients have shown conflicting results.Recently,some researchers reported that blood A?42 concentration is affected by factors including obesity,blood lipids,and blood pressure.In order to reveal the correlation between obesity,obesity-related factors and blood A?42 levels,as well as the impact on AD patients A?42 levels,this population-based cross-sectional study tries to study the association of obesity,blood lipid,blood pressure factors and serum A?42 and analyzed the diagnostic ability of the serum A?42 levels between AD patients and healthy people after correcting other related factors.Materials and Methods: This study recruited 1261 healthy volunteers who had physical examinations at the Sichuan Provincial People's Hospital Health Checkup Center from January 2019 to December 2019,and recruited 22 AD patients at the Sichuan Provincial People's Hospital and Zigong First People's Hospital from October 2019 to January 2020.The serum A?42 concentration of the subjects was tested by enzyme-linked immunosorbent assay,and used statistical methods to assess its association between the body mass index(BMI),total cholesterol(TC),triglycerides(TG),high-density lipoprotein cholesterol(HDL-c),low-density lipoprotein cholesterol(LDL-c),systolic blood pressure(SBP)and diastolic blood pressure(DBP),and the diagnostic efficacy of A?42.Results: In healthy people,There were significant correlation among serum A?42 level and BMI(? = 0.316,p <0.001),high-density lipoprotein cholesterol cholesterol(? =-0.091,p = 0.034)and diastolic blood pressure(? = 0.108,p = 0.005);after correcting the influencing factors,serum A?42 has a certain diagnostic effect on Alzheimer's disease,the area under the ROC curve is 0.788,sensitivity = 88.5%,specificity = 83.3%.Conclusion: This study found that the serum A?42 level in Chinese population is affected by BMI,HDL-c,DBP,and serum A?42 has the potential as a diagnostic marker for AD after correcting related influencing factors.
