Medication Law Analysis Of Osteoporosis By Using Data Excavation And The Mechanism Of Tianchang Capsules For Ovariectomy-induced Osteoporosis In Rats

Objective:To explore the prescription of traditional Chinese medicine for Chen qiu clinical treatment of osteoporosis(OP)by data excavation to analyze the common drugs and prescription rules,for summaring the academic experence of treatment OP,and to lay a theoretical foundation for the formulation of Tianchang Capsule.To explore the mechanism of Tianchang Capsule on ovariectomized rats to play an anti-osteoporosis for further study.Method:1.Through the outpatient system of Sichuan traditional Chinese Medicine Hospital collected the prescriptions of Chen qiu to treat osteoporosis from September 2016 to September 2019.The database is established by excel,and the data mining analysis is carried out with SPASS 23.02.Divided 60 female SD rats into 6 groups after ovariectomy or sham operation,sham operation group(Sham,n=10),model control group(OVX,n=10),positive control group(OVX+AL,n=10),Tianchang capsule low dose group(OVX+LTC,n=10),Tianchang capsule medium dose group(OVX+MTC,n=10)and Tianchang capsule high dose group(OVX+HTC,n=10).According to the set dosage,after 12 weeks,then bone mineral density,bone histopathology and uterine coefficient were measured,enzyme-linked immunosorbent assay(ELIS A)was used to measure the serum E2、OPG and RANKL.Results :1.A total of 114 prescriptions were included in this study,a total of 115 Chinese medicine,drug frequency: a total frequency of 2085 times.The first15 kinds of traditional Chinese in the accumulative frequency are Astragalus Membranaceus、Angelica sinensis、Codonopsis pilosula、Licorice Roots Northwe st Origin、Atractylodis Macrocephalae、 loranthus parasiticus、herba epimedii、fructus aurantii、Poria cocos、concha ostreae、fructus psoraleae、radix dipsaci、Eucommia、Radix Rehmanniae Praeparata、Bupleurum.Main classification of drugs: tonifying deficiency(53.9%),activating blood circulation and removingblood stasis(4.41%),heat-clearing(6.52%),regulating qi(5.32%),calming the liver wind(6.95%),clearing damp and promoting diuresis(6.52%),diaphoretic drug(5.27%),dispelling wind-damp(5.56%).Cluster analysis of three groups of prescriptions and factor analysis of seven related factors.2.Compared with the sham-operated group,the bone mineral density(BMD)of OVX group was decreased(P<0.01).Compared with OVX group,the BMD of the OVX+AL,OVX+TCLand OVX+TCM group was increased(P<0.05).Compared with OVX group,each dose group of Tianchang Capsule and positive control group were significantly improved the pathological changes of bone tissue morphology.The ratio of RANKL to OPG in the positive control group and Tianchang capsule low dose group were significantly increased(P<0.05),comparing with the OVX group.The uterine coefficient and serum E2 levels were not statistically significant compared with OVX in OVX+AL、OVX+TCL、OVX+TCM、OVX+TCH(P >0.05).Conclusion:1.The core drugs of Chen qiu to treat OP are Astragalus membranaceus、Angelica sinensis、Codonopsis pilosula、Licorice Roots Northwest Origin、Atrac-tylodis Macrocephalae、 loranthus parasiticus、herba epimedii、fructus aurantii Poria cocos、concha ostreae、fructus psoraleae、radix dipsaci、Eucommia、Radi x Rehmanniae Praeparata、Bupleurum.Pay attention to replenish qi and nourish blood,replenish liver and kidney,and promote blood circulation,eliminating dampness are added at appropriate time.2.Tianchang capsule can play an anti-OP role by regulating OPG/RANKL/RANK signaling pathway.
