Objective:To explore the prevention of resveratrol for osteoporosis in ovarian rats by adjusting the activity of antioxidant enzymes.Methods:According to the randomized grouping principles,30 clean grade SD rats were randomly divided into five groups (n=6 for each) as following details:the false surgical group(SHAM), the ovariectomized group (OVX), low dose of RES treatment group (RL), moderate doses of RES treatment group (RM), high doses of RES treatment group (RH). Each group of rats was established to postmenopausal osteoporosis model(except the SHAM) by surgical removing bilateral ovaries. At one week after the bilateral ovariectomy operation completely, RES was preventive treatment and weighing was weighted weekly. After 12 weeks continuous intervention,got blood and serum biochemical indicators were detected, after all animals were sacrificed, collected their bone tissue quickly and uterine wet weight were collected. Dual energy X-ray absorptiometry was used to determinate the rats bone density.HE staining was used to observe bone tissue morphological changes under the light microscope, and the reverse transcription RCR (RT-PCR) was used to detect the mRNA expressions of antioxidant enzymes in bone tissue y-GCSh HO-1 CAT.Results:1.Compared with the SHAM model group, serum biochemical indicators in OVX were changed as details:Ca2+, IP, alkaline phosphatase (ALP) and tartrate resistant acid phosphatase-5b (TRACP-5b) were significantly increased, estrogen levels were significantly decreased and uterine wet weight were decreased; HE staining and the bone density tests showed that the bone tissue was in osteoporosis status, the bone density was reduced obviously; the antioxidant enzymes as γ-GCSh,HO-1 and CAT were significantly down-regulated in mRNA expression.2. Compared with the OVX group, serum biochemical indicators in group treated by(ALP) and tartrate resistant acid phosphatase-5b (TRACP-5b) were significantly decreased, estrogen levels were significantly increased; HE staining and the bone density tests showed that the bone tissue was improved in osteoporosis status, bone density was increased. the antioxidant enzymes as y-GCSh,HO-1 and CAT were significantly up-regulated in mRNA expression.Conclusion:1.RES has certain inhibitory effect to metratrophia of ovariectomized rats. 2.RES can effectively reduce the elevated serum ALP, TRACP-5b, Ca2+, and IP, to improve the state of high conversion to ovarian rats.3.RES in 20 to 40 mg/(kg·d) dose range can effectively restrain to ovarian rats bone loss, improve to the ovaries rat bone density, its mechanism may be related to its have estrogen-like activity.4.RES can enhance the activity of the antioxidant enzymes to ovarian rats bone y-GCS, HO-1 and CAT, improve oxidative stress state.And it have certain prevention of osteoporosis in rats.
