The Curative Effect And Mechanism Of Tianchang Capsule For Ovariectomy Induced Osteoporosis In Rats

Objective: This article performed a model of osteoporosis rats using the method of ovariectomy and observed efficacy of Tianchang Capsule on treatment of osteoporosis rats.Comparing the effect with positive drug to explore the mechanism of Tianchang Capsule in osteoporosis.Methods: Female SD rats were randomly assigned to 6 groups(10 in each group): control(sham-treated),OVX(by ovariectomy),OVX+AL(by ovariectomy,received alendronate sodium),OVX+LTC(by ovariectomy,received low dosage Tianchang Capsule),OVX+MTC(by ovariectomy,received middle dosage Tianchang Capsule),OVX+HTC(by ovariectomy,received high dosage Tianchang Capsule).The sham-treated group remove the near fat part of the ovary,the other groups remove the bilateral ovaries.After ovariectomy operation,alendronate or Tian Chang capsule were given for 12 weeks,then the rats were executed and bone mineral density(BMD),bone resorption and bone formation indexes and femoral pathological were determined.Results:Bone mineral density(BMD):Compared with Shame-treated group,the BMD of OVX group had a significant decrease(P < 0.01).OVX+AL,OVX+LTC and OVX+MTC groups increased in BMD comparing with OVX.Bone formation parameters: Compared with the Shame-treated group,model control group of bone formation index(BALP and BGP)were significantly increased(P < 0.01).The BALP level have a statistical significance compared with OVX only in OVX+AL group(P<0.05).The BGP level have a statistical significance compared with OVX only in OVX+LTC group(P<0.05).Bone resorption parameters: Compared with the Shame-treated group,model control group of bone resorption index(DPD and TRACP)were increased(P < 0.01 or P<0.05).The DPD level have a significantly statistical significance compared with OVX in OVX+AL and OVX+LTC groups(P<0.01).The TRACP level all have statistical significance between OVX and four treatment groups.Histomorphological Results: OVX group were evident damaged in trabecular bone.The trabecular bone in four treatment group were improved in varying degrees.Conclusion: Tianchang capsule has good curative effect in treatment of ovariotomy osteoporosis rats,which play roles in enhance the level of bone mineral density and restrain excessive active bone resorption,and also improve the trabecular bone damage.
