Objectives To investigate the associations between occupational harmful factors,including high temperature,noise,dust,carbon monoxide(CO),shift work,rhythm-related genes polymorphisms and their interactions with obesity in steel workers.Methods A cross-sectional study was adopted to analyze the data with 7664 workers of the HBIS GROUP TANGSTEEL COMPANY based on the health effect cohort study of the occupational population in the Beijing-Tianjin-Hebei region established by the research group,after relevant information was collected by questionnaire and physical examination,restricted cubic spline(RCS)model was used to analyze the dose-response relationship between the occupational harmful factors and obesity,then the multi-factor logistic regression model was used to analyze the relationship between the occupational harmful factors and their interactions with obesity.In the subsequent study,394 obese patients and 581 controls were randomly selected from the 7664 workers with complete data to analyze the relationship between rhythm-related genes polymorphisms and obesity,with a case-controll study.Four eligible circadian-related genes were selected,the genotype of target gene locus was determined by polymerase chain reaction(PCR)and limiting fragment length polymorphism,the influence of the interactions between genes and genes,genes and environment on obesity was analyzed by multi-factor logistic regression model and generalized multi-factor dimensionality reduction model(GMDR).Results 1 7664 workers were selected into the study,including 7031 males and 633 females.The prevalence of obesity in the study population,male,and female were: 22.26%(1706/7664),23.13%(1626/7031),and 12.64%(80/633),respectively.The average body mass index(BMI)was 25.2 kg/m2;2 The RCS models showed that compared with occupational hazards cumulative exposure measurement(CEM)=0,there was Non-linear dose-response relationship between WSI,high temperature,noise,dust,CO CEM and obesity(P<0.05 for overall association test,P<0.05 for nonlinear test)after adjusting for age gender,education level,monthly income per capita,smoking,drinking,dietary,sleep duration,physical activity,high temperature,noise,dust,and CO CEM;3 The multi-factor logistic analysis showed that the prevalence of obesity reduced in the 30~and 40~years old workers,compared with the referrent(20~year old),OR(95%CI)=0.721(0.540~0.963)and 0.552(0.400~0.762),respectively.The likelihood of obesity reduced when the CO CEM was more than 52.24 mg/m3·year,compared with the referent(CO CEM=0)(OR=0.673,95% CI:0.393~0.953);4 Interaction was found between WSI and age,smoking,noise CEM,high temperature and age,gender,sleep duration,CO CEM,noise CEM and age,smoking,dust CEM,dust CEM and dietary,education level,based on the multiplication model.5 The risk of obesity increased by 43%,with the genotype of the RORA gene rs8034880 locus is GG compared to the AA and AG(OR=1.43,95%CI:1.05~1.94);As for the MTNR1 A gene rs 2119882 locus,workers who carried the CC genotype reduced the risk of Obesity compared to the TT and TC(OR=0.60,95%CI: 0.40~0.89),while the TC increased the risk of obesity by 44%,compared to the TT and TC(OR=1.44,95%CI: 1.10~1.87).6 The association of CLOCK gene rs1801260 locus polymorphism and PER2 gene rs2304672 locus polymorphism with obesity was not found to be statistically significant,(P>0.05);7 There is an interaction between CLOCK gene rs1801260 locus and MTRN1 A gene rs2119882 locus based on multiplicative model,(Pinteraction=0.020);8 There is no interaction between rhythm-related genes and occupational harmful factors based on multiplicative model(Pinteraction>0.05).Conclusions 1 There is a nonlinear dose response relationship between WSI,high temperature CEM,noise CEM,dust CEM,CO CEM and Obesity;2 RORA gene rs8034880 locus and MTRN1 A gene rs2119882 locus gene polymorphism is associated with Obesity;3 There is an interaction between MTRN1 A gene rs2119882 locus and CLOCK gene rs1801260 locus based on multiplicative model.Figure9;Table30;Reference 100...
