Objective:This study focuses on discussing circadian clock gene Per3 polymorphism,DNA methylation profile of promoter region,environmental risk factors,and the relationship between the interaction and breast cance,which provides certain theoretical basis for the pathogenesis of breast cancer,psychological intervention,and prognosis.Methods:Collecting 134 breast cancer patients and adopted 1:1 matching principle to collect 134 he althy people and conducting a popular questionnaire survey about social environmental factors of self-designed general demographic questionnaire,the Pittsburgh sleep quality index scale(PSQI),concise occupational stress questionnaire,trait coping style scale and the social support scale.We use polymerase-chain reaction(PCR)for the amplification of Per3 gene in peripheral blood,direct sequencing for SNP detection and cloning sequencing for the gene methylation.Results:(1)Analysis of the social environment factors in the case control group showed the total score of the case group of sleep quality,sleep duration,sleep disorder,daytime dysfunction dimension and PSQI was higher than the control group,and the difference was statistically significant(P<0.05),the social support of case group was lower than that of the control group and the difference was statistically significant(P<0.05),and total score of other dimensions of occupational stress had no difference in both group(P>0.05), the negative coping scores of trait coping style scale in the case group were higher than those in the control group,the difference was statistically significant(P<0.05),total score of ubjective support,support utilizationsocial support in case group were lower than those of control group,the difference was statistically significant(P<0.05);multi factor logistic regression analysis showed that social risk factors of breast cancer were sleep time,sleep efficiency,sleep disorders,social support,positive coping,objective support and support utilization(P<0.05);(2)Analysis of Per3 gene polymorphism in case control group suggested genotype of rs2797687 and rs2794664 were statistically signific- ant differencesin the two groups(P<0.05),but not rs228730,rs571514100(P>0.05); interaction analysis showed that the Per3 rs2797687 T/T had a positive effect on the interaction of sleep quality(OR=1.313,P<0.01),and the combined effects may increase the risk of breast cancer by 1.313 times;Per3 rs2797687 T/T and G/T had a negative multiplicative interaction with social support(OR=0.948,P<0.01;OR=0.959,P=0.036),the joint action may reduce the risk of breast cancer by 5.2%and 4.1%,respectively.Per3 rs2794664 A/A and sleep quality had a positive multiplicative interaction(OR=1.29, P<0.01), which may increase the risk of breast cancer by 1.29 times;Per3 rs2794664 A/A and social support have a negative multiplicative interaction(OR=0.951,P<0.01),which may cause the reduction of breast cancer risk to 4.9%;Per3 rs228730 G/A and negative coping had a positive multiplicative interaction(OR=1.366,P=0.036),and the common role may increase the risk of breast cancer by 1.366 times;Per3 rs228730 G/A genotype and social support had a negative multiplicative interaction(OR=0.82,P=0.044),and its interaction may reduce the risk of breast cancer by 18%.(3)Methylation level of CpG island in promoter region of Per3 gene showed that the total methylation rate of Per3 gene in case group was higher than that in control group,and the difference was statistically significant(P<0.05);binarylogistic regression analysis showed that the rs2797687 locus mutant individuals were more likely to have higher methylation compared to the wild-type individuals(OR=1.960),and there was no statistical significance(P>0.05).The correlation between Per3 rs228730 and rs2794664 SNP and CpG island of Per3 gene was not significant(OR=0.964,0.889,P>0.05).Conclusion:Sleep quality,occupation stress,coping style and social support are the social risk factors of breast cancer,there was interaction between the Per3 gene rs2797687,rs2794664 and rs228730 mutation and the social environment factor,the independent risk factors of CpG island methylation was Per3 gene single nucleotide polymorphism,detecting Gene polymorphism and methylation of Per3 gene may have made sense for the prevention and prognosis of breast cancer.
