Morphological Observation Of Cantharidin Cream Intrauterine Administration On Endometrial Hyperplasia Model Rats

Objective: In order to provide new ideas and methods for clinical treatment of AUB-O,the weight,wet weight of uterus,index of uterus,morphological changes of endometrium under light and electron microscope were observed in rats with different concentration of cantharidin cream under different administration time.Methods: 42 healthy adult female SD rats were randomLy divided into 4 groups: group A:sham operation group,group B: model group,group C: low dose group,group D: high dose group.In group A,the rats were operated only by abdominal switch.The other groups of rats underwent bilateral ovariectomy,followed by intramuscular injection of estradiol benzoate to create endometrial hyperplasia models.After the models were established successfully,0.3mL normal saline was injected into the bilateral uterine cavities of group B,0.3mL cantharidin cream of low concentration was injected into the bilateral uterine cavities of group C,and 0.3mL cantharidin cream of high concentration was injected into the bilateral uterine cavities of Group D.After medication for 3 days,half of the rats in each group were randomLy selected and killed.After medication for 5 days,all the remaining rats were taken out of uterus and killed.The weight and wet weight of uterus were measured,and the index of uterus was calculated.The changes of microstructure and ultrastructure of uterus were observed under light and electron microscope.Results:1.The rats in each group were generally in good condition with no significant difference in final body weight(P > 0.05).2.There were significant differences in wet weight and index of uterus between group B and group A(P<0.05);compared with group B,there were significant differences in wet weight and index of uterus in group C and group D(P<0.05);there were significant differences in wet weight and index of uterus between group C and group D(P<0.05)after medication for 3 days;there were significant differences in wet weight and index of uterus between group C and group D(P<0.05)after medication for 5 days;there was no significant difference in wet weight and index of uterus between 3 days,medication and 5days,medication in group C(P>0.05);there were significant differences in wet weight(P<0.05)and index of uterus(P<0.05)between 3 days,medication and 5 days,medication in group D.3.There was a significant difference in endometrial thickness between group B andgroup A(P<0.05);endometrial thickness of group C and group D were significantly different compared with group B(P<0.05);after medication for 3 days,there was a significant difference in endometrial thickness between group C and group D(P<0.05);after medication for 5days,there was no significant difference in endometrial thickness between group C and group D(P > 0.05);in group C and group D,there was a significant difference in endometrial thickness between 3 days,medication and 5 days,medication(P<0.05).4.Group A: the epithelial cells of the mucosa were monolayer columnar,the glands were sparse and scattered,and the epithelial cells of the glands were cubic,arranged in monolayer or pseudostratified.Group B: the epithelial cells of mucous membrane were in high columnar shape,the number of glands increased,some glands expanded into sacs,the epithelial cells of glands proliferated,in high columnar shape,and arranged in pseudostratified arrangement.Group C: after medication for 3 days,mucosal epithelium partial lost,visible inflammatory cell infiltration,glandular necrosis,glandular cell exfoliation,inflammatory cell infiltration can be seen in the glandular cavity.Compared with 3 days,medication,the infiltration of inflammatory cells and the degree of edema in the whole layer of uterus decreased,the intima thickened and the number of glands increased,blood vessels were abundant in those who got a 5-days,medication.Group D:after medication for 3 days,all of mucosa epithelium lost,gland disappeared,a large number of inflammatory cells infiltrated,muscle layer was edematous.Compared with 3days,medication,the infiltration of inflammatory cells and the degree of edema in the whole layer of uterus were reduced,the intima was thickened,the number of glands was increased,and blood vessels were abundant in those who got a 5-days,medication.5.Group A: normal structural organelles were found in cytoplasm.Group B: the secretory cells were pseudostratified and high columnar,with more mitochondria in the cytoplasm,abundant rough endoplasmic reticulum and Golgi complex,larger nucleus,more chromatin and obvious nucleolus.Group C: after medication for 3 days,secretory cells were monolayer and low columnar.Compared with group B,there were less mitochondria in cytoplasm,less rough endoplasmic reticulum,less Golgi complex,shrinking nuclear membrane,less chromatin and disappeared nucleolus.Compared with 3days,medication,those who got a 5-days,medication had more mitochondria,less rough endoplasmic reticulum expansion,stable nuclear membrane and multiple nucleoli.Group D: after medication for 3 days,secretory cells were monolayer cubic.Compared with group B,mitochondria in cytoplasm were significantly reduced,and autophagic bodies with double membrane structure were observed,rough endoplasmic reticulum was reducedand expanded obviously,Golgi complex was reduced,nuclear morphology was basically normal,chromatin was reduced,and nucleolus disappeared.Compared with 3 days,medication,the expansion degree of endoplasmic reticulum was reduced,necrotic cells were seen to be engulfed,chromatin in nucleus increased,and more than one nucleoli appeared in those who got a 5-days,medication.Conclusion:1.Cantharidin cream has no effect on general condition and body weight of rats.2.The effect of cantharidin cream on rat endometrium is dose-dependent.3.After the action of cantharidin cream for a period of time,the endometrium can grow again.It can be seen that the medicine does not cause irreversible damage to the basement of endometrium.However,even at low concentration,the whole layer of the uterus is more edematous and inflamed,so it can be considered to use a smaller dose than the low-dose group,repeated use of drugs for many times.4.Cantharidin cream may play a role by inducing degeneration,necrosis or autophagy of endometrial cells in rats.
