| objectiveThe purpose of this study was to investigate the distribution abundance of high-variant long non-coding RNA(LncRNA)in the peripheral blood of the lung cancer of the Xuanwei family and its relationship with clinical characteristics,and to explore its effect on the genetic susceptibility of lung cancer.Methods(1)In the early stage of the research group,the top ten LncRNAs with high-frequency mutations in Xuanwei familial lung cancer were screened by Genome Wide Association Studies(GWAS),and primers were designed for the high-frequency mutation sites.(2)Detecting the distribution abundance of target LncRNA in peripheral blood of lung cancer patients and its relationship with clinical features of lung cancer by qRT-PCR technology.(3)To explore the sensitivity of the cell line to the carcinogen Benzopyrene(Bap)after LncRNA silencing or overexpression through cell function pre-experiment.Results(1)In the correlation analysis of the target LncRNA and clinicopathological indicators of all patients:the blood cell LINC00343 showed a significant correlation with the gender of the patient(p=0.047),and the male patients whose expression level was higher than the average were twice the female patients(Response to the gender correlation of LINC00343);blood cell LINC00343 showed a correlation with the smoking history of patients(p=0.036),blood cell LINC00343 was more likely to be highly expressed in the smoking patient population;plasma LINC00343 was significantly biased in the patient population older than 55 years High(p=0.03)(response LINC00343 showed an upward trend in elderly patients);plasma LINC00343 was significantly increased in patients with stage Ⅲ-Ⅳ lung cancer(p=0.01)(responding to the possible cancer-promoting effects of LINC00343);blood cells LINC00433 It showed a significant correlation with multiple nodules in the lungs(p=0.017).In patients without multiple nodules in the lungs,the blood cell LINC00433 was higher(reporting the possible tumor suppressor protective effect of Linc00433).(2)In the correlation analysis of target LncRNA and clinicopathological indicators of patients with adenocarcinoma squamous cell carcinoma:the blood cell LINC00343 showed a significant correlation with the gender of the patient(p=0.047),male patients whose expression level was higher than the average were two female patients Times(response to the gender correlation of LINC00343);blood cells LINC00343 showed a correlation with the smoking history of patients(p=0.036),blood cells LINC00343 were more likely to be highly expressed in smoking patient groups;plasma LINC00343 was more than 55 years old in patient groups Moderately high(p=0.03)(response LINC00343 showed an upward trend in elderly patients);plasma LINC00343 was significantly increased in patients with stage Ⅲ-Ⅳ lung cancer(p=0.01)(responsive to the possible cancer-promoting effects of LINC00343);The blood cell LINC00433 showed a significant correlation with multiple nodules in the lungs(p=0.017),and the blood cell LINC00433 was higher in patients without multiple nodules in the lungs(in response to the possible tumor suppressive protective effect of LINC00433)(3)Cell pre-experiment found that LINC00433 and LINC00842 exhibited tumor suppressor protection under certain conditions.LINC00343 exhibits cancer-promoting function under certain conditions.ConclusionBlood cells LINC00343,plasma LINC00343,blood cells LINC00433,blood cells LINC00842 may become potential new biological markers for diagnosis and treatment of lung cancer. |
