| Objective:Lung cancer rates in Xuanwei County, Yunnan Province, are among the world’s highest, which have genetic characteristics with a high incidence in women and familial aggregation. We analysis genome-wide CNVs with xuanwei lung cancer by high-reslution CGH arrays, trying to screened out a set of CNVs regions or genes as molecular markers, related Xuanwei lung cancer.Methods:1. we performed analysis of CNVs using Nimblegen Human CGH Array on8patients’tumor and adjacent tissues. CNVs data compare with non-small cell lung cancer database, screened out a set of candidate CNVs regions or genes for xuanwei lung caner;2.16of obtain CNVs regions and22genes were verified in43cases using SYBR Green real-time PCR;3. Correlation analysis of candidate genes copy number changes with clinical data, and characteristics of candidate genes function, pathway^biological process and tissue distribution online. Comprehensive understanding of the mechanism of candidate genes in Xuanwei lung cancer.Results:1. We identified16CNVs regions,6regions for copy number gain and9regions for copy number loss.2. Compare with CNVs database of non-small cell lung caner,9overlapping chromosomal regions (5p15.33ã€5p15.33-pl5.32.8q22.1ã€5q35.2-q35.3.7q21.11-q21.3ã€11q24.2-q24.3ã€18q21.1-q21.2,2q31.1-q32.3and19q13.2-q13.31) were identified by co-localizing disease loci for other tumor types.4novel regions (5q31.1-q33.1ã€12p13.32-p13.3ã€Xp11.23-p11.l and4p16.3) have different trends with the database of CNVs.Interestingly, the Deletion in Xp11.3-p11.23regions is a the newly discovered related by lung cancer.3.22selected candidate genes were verified by real-time PCR and they were all verified the array result (p>0.05).4.We identified22genes associated xuanwei lung cancer,2new genes(FGF6and FGF23) were not reported have association with lung cancer before. Candidate genes copy number changes were relevant with clinical data, and we also identified relationship networks between the candidate genes, their functions and biological processes or pathways play an important role in the occurrence and development of tumors.Conclusion:Our study suggested that CNVs might be potentially important for the Xuanwei lung cancer,they play an important role in the occurrence and development of Xuanwei lung cancer, and they might be used as a genetic marker to locate with Xuanwei population. |
