Purpose: To investigate the effect of electroacupuncture stimulation on the expression of NGF and Trk A in damaged spinal cord segments in rats with spinal cord injury.Methods: The model of spinal cord injury was replicated by the rat spinal cord semi-transverse method.On the 3rd day after the successful modeling,the rats were randomly divided into the Dumai acupuncture group,the foot-yangming gastric acupuncture group and the Injury control group,respectively using electric acupuncture to stimulate the Dumai acupuncture point,the foot-yangming gastric acupuncture point and the non-use of electric acupuncture.The neurological function scores of the rats in each group were evaluated by BBB scoring method.The rats were sacrificed 14 days after treatment.The damaged spinal cord segments of the rats in each group were collected and the tissue damage was observed by HE staining.Immunohistochemical staining was used to compare the expression of NGF and Trk A positive cells in each group.m RNA expression of NGF and Trk A in each group was compared by q RT-PCR.Results: There was no significant difference in BBB function score of each group before treatment.BBB score at 1 week of treatment and 2weeks of treatment: Yangming group > Dumai group > Injury control group;HE staining showed that the spinal cord injury in each group was consistent with the functional score.Immunohistochemical staining results showed that the expression of NGF plays in the seventh day of electric acupuncture intervention: Yang Ming group > Dumai group >Injury control group,Electric acupuncture intervention day 14: Yang Ming group > Dumai group > Injury control group,Yang Ming group expression quantity: 7 days > 14 days,Du meridian acupuncture point group expression quantity: 7 days > 14 days,The control group expression quantity: 7 days > 14 days,the expression of Trk A situation in the seventh day: Yang Ming group > Dumai group > Injury control group,the 14 th day: Yang Ming group > Dumai group > Injury control group,Yang Ming group expression quantity: 7 days > 14 days,Du meridian acupuncture point group expression quantity: 7 days > 14 days,the expression level of the injury control group was as follows: 7 days > 14 days.q RT-PCR results showed that the expression of NGF plays in the seventh day of electric acupuncture intervention: Yang Ming group >Dumai group > Injury control group,electric acupuncture intervention day 14: Yang Ming group > Dumai group > Injury control group,Yang Ming group expression quantity: 7 days > 14 days,Du meridian acupuncture point group expression quantity: 7 days > 14 days,the expression level of the injury control group was as follows: 7 days > 14 days,the expression of Trk A situation in the seventh day: Yang Ming group > Dumai group > Injury control group,the 14 th day: Yang Ming group > Dumai group > Injury control group,Yang Ming group expression quantity: 7 days > 14 days,Du meridian acupuncture point group expression quantity: 7 days > 14 days,the expression level of the injury control group was as follows: 7 days > 14 days.Conclusion: By increasing the protein and m RNA expression of NGF and Trk A in spinal cord injury segment,electroacupuncture stimulation can achieve the recovery and intervention of SCI dysfunction. |
