Yunpi Rougan Formula is the experience gained by Professor Tian Yaozhou through many years of clinical practice,summarized the formula that takes the liver and spleen as the main method.It has the effects of softening the liver and spleen,regulating qi and defecation,and received good clinical effect in the clinical treatment of constipation-Predominant irritable bowel syndrome(IBS-C).Based on the previous clinical and experimental results of the research group,this topic further explores the theoretical basis and mechanism of the effect of Yunpi Rougan Formula.Part 1:Theoretical study:From the perspective of modem medicine and traditional Chinese medicine,this article summarizes and analyzes the etiology,pathogenesis,diagnosis and treatment of IBS-C.The research status indicates that modern medicine has made certain achievements in the research and understanding of IBS-C-related causes and diagnosis and treatment.Compared with western medicine treatment,traditional Chinese medicine treatment of IBS-C can alleviate the discomfort and recurrence rate of patients.Lower,has certain advantages in terms of treatment.In addition,the theoretical mechanism of Yunpi Rougan Recipe’s therapeutic effect on IBS-C is explained in terms of the drug composition and its significance,and the pharmacological effects of traditional Chinese medicine.Part 2:Experimental researchOBJECTIVE:To observe the effects of Yunpi Rougan Recipe on the expression of motilin(MOT),somatostatin(SS)and SCF/c-kit signaling pathway in serum and colon tissue of IBS-C rats To explore the effect mechanism of Yunpi Rougan Recipe on the improvement of intestinal motility in IBS-C rats.METHODS:Twelve wistar rats were selected as the normal group by random number table method,and the rest of the rats were established by ice water gavage to establish the IBS-C model.After successful modeling,the rats were divided into IBS-C model group,mosapril group and low,medium and high dose groups with 12 rats in each group according to the random number table method,.The normal group and the IBS-C model group were given equal volume of distilled water,and the mosapride group was given 1.35 mg/kg/d mosapride suspension,the low,medium and high dose groups were given 12.2g,24.4g and 48.8g/kg/d,respectively.Calculate the number of fecal particles and the water content of the feces,and measure the intestinal propulsion rate with the ink propulsion test.Enzyme-linked immunosorbent assay(ELISA)and immunohistochemistry to determine the expression of MOT and SS in rat serum and colon tissue.Immunohistochemistry and Q-PCR were used to detect the expression of SCF,c-kit protein and mRNA in colon tissues.RESULTS:① Compared with the normal group,the number of fecal particles,fecal water content,and intestinal propulsion rate in the IBS-C model group were significantly reduced;compared with the IBS-C model group,fecal particles,fecal water content,and intestinal propulsion rate in the infusion medicine groups were significantly reduced.Both are elevated.② Compared with the normal group,the expression of MOT in serum and colon tissues of the IBS-C model group decreased(P<0.05),and the expression of SS increased(P<0.05);compared with the IBS-C model group,the serum and colon tissues of the infusion medicine groups,the expression of MOT increased and the expression of SS decreased,among which the mosapride group and the Chinese medicine medium and high dose groups were statistically significant(P<0.05).③ Compared with the normal group,the expression levels of SCF,c-kit protein and mRNA in the colon tissue of the IBS-C model group were significantly reduced(P<0.01);comp ared with the IBS-C model group,the SCF and c-kit proteins in the colon tissue of the infusion medicine groups and mRNA expression levels increased,except that the expression level of SCF protein in the low-dose group of traditional Chinese medicine was not statistically significant,and all were statistically significant(P<0.01~0.05)CONCLUSION:①Yunpi Rougan Formula can significantly improve the defecation and stool characteristics of IBS-C rats and increase the rate of bowel movement.②After drug intervention,the expression of MOT in serum and colon tissues in the treatment group increased,and the expression of SS decreased,indicating that one of the mechanisms of Yunpi Rougan Formula in the treatment of IBS-C may be to enhance MOT expression and inhibit SS expression and regulate the body Abnormal expression of gastrointestinal hormone levels,which improves bowel movements and promotes bowel movements,and improves IBS-C symptoms.③After drug intervention,the expression of SCF,c-kit protein and genes in the colon tissue of the treatment group was enhanced.One of the mechanisms for Yunpi Rougan Formula to treat IBS-C may be to improve the intestinal tract by increasing the expression of SCF/c-kit signaling system.Power to achieve the effect of treating IBS-C.
