The Expression Of SHIP2 And Its Clinical Significance In Breast Carcinoma

Objective: To investigate the expression of SHIP2 protein in breast carcinoma tissues,normal tissues adjacent to breast carcinoma and the lymph nodes of breast carcinoma patients,to approach the potential function of SHIP2 as well as its clinical significance in the development of breast carcinoma,and the correlation of lymph node metastasis of breast carcinoma.Method: Employ Western blot and qPCR to test the expression of SHIP2 protein and SHIP2 mRNA in 60 cases of breast carcinoma and normal tissues adjacent to it.Use immune histochemistry to test the expression of SHIP2 protein in 50 cases of the lymph nodes in lymph node negative transfer groups(Group A),10 cases of the metastatic lymph nodes in lymph node positive transfer groups(Group B).Results: Western blot shows the relative expression of SHIP2 protein in breast carcinoma tissues and normal tissues adjacent to breast carcinoma,is(0.708±0.135)and(0.3±0.057)respectively.qPCR shows that the relative expression of SHIP2 mRNA in breast carcinoma tissues and normal tissues adjacent to breast carcinoma,is(1±0.002)and(0.18±0.208)respectively.The expression of SHIP2 protein in breast carcinoma tissues is far higher than that in normal tissues adjacent to breast carcinoma(P<0.05);the expression of SHIP2 mRNA in breast carcinoma tissues is far higher than that in normal tissues adjacent to breast carcinoma(P<0.05);immune histochemistry indicates the positive rate of SHIP2 in Group A and Group B is 18%(9/50)and 70%(7/10),the positive rate of SHIP2 in the lymph nodes in Group B is higher than that in Group A,the difference was statistically significant.Conclusion: The over expression of SHIP2 is closely related to breast carcinoma,which is likely to be the target of preventing and controlling breast carcinoma.The over expression of SHIP2 is likely possess the positive correlation with Lymph node metastasis of breast carcinoma.
