Objective:To assess the expression and significance of the apoptosis-related gene Bag-1(Bcl-2associated athanogenel)ã€Bcl-2(B cell lymphoma/leukemia gene2)〠Bax(Bcl-2associated X protein) in breast cancer and to analyze the association of its expression with clinicopathological characteristics and prognosis of breast cancer patients.Methods:We choose85invasive ductal carcinoma(IDC) of breast and its tumor-adjacent normal tissues,75ductal carcinoma in situ(DCIS) of breast and its tumor-adjacent normal tissues from the affiliated cancer hospital of tianjin medical university. S-P immunohistochemistry method was used to assess the expression of Bag-1ã€Bcl-2〠Bax in breast tumor and tumor-adjacent normal tissues.Results:1. Immunohistochemical staining results revealed that the positive expression of Bag-1showed cytoplasm and nucleus staining, Bcl-2ã€Bax showed cytoplasm staining.The expression of Bag-1in IDC of breast was (-)6(7.1%)ã€(+)21(24.7%)ã€(++)28(32.9%)ã€(+++)30(35.3%), higher than in tumor-adjacent normal tissues was (-)35(41.2%)ã€(+)34(40.0%)ã€(++)16(18.8%)ã€(+++)0(0%), the difference has statistics meaning (P1=4.06x10-12). That was the similar trend in DCIS of breast and its tumor-adjacent normal tissues (P2=2.17×10-4). And the expression of Bag-1in IDC of breast was higher than in DCIS of breast (P3=5.71×10-5)The expression of Bcl-2in IDC of breast was (-)18(21.2%)ã€(+)24(28.2%)ã€(++)27(31.8%)ã€(+++)16(18.8%), higher than in tumor-adjacent normal tissues was (-)35(41.2%)ã€(+)33(38.8%)ã€(++)17(20.0%)ã€(+++)0(0%), the difference has statistics meaning (P1=8.05×10-6). That was the similar trend in DCIS of breast and its tumor-adjacent normal tissues (P2=0.026). And the expression of Bcl-2in IDC of breast was higher than in DCIS of breast (P3=0.040)The expression of Bax in IDC of breast and its tumor-adjacent normal tissues, in DCIS of breast and its tumor-adjacent normal tissues, in IDC of breast and DCIS of breast was no statistics meaning.Bag-1ã€Bcl-2ã€Bax was high positive correlation in IDC of breast and DCIS of breast.2. The association between the expression of Bag-1ã€Bcl-2ã€Bax and clinicopathological characteristics in IDC of breast.The expression of Bag-1was correlated with tumor size, and was significantly higher in tumor>2cm than in tumor≤2cm(P=0.001), but was not correlated with lymph node metastasis, clinical staging, expression of estrogen receptor(ER) and progestogen receptor(PR) in IDC. The expression of Bcl-2and Bax was not correlated with tumor size, lymph node metastasis, clinical staging, expression of ER and PR in IDC.3. The association between the expression of Bag-1ã€Bcl-2ã€Bax and prognosis in IDC of breast.The Log-rank test and the Cox regression single factor analysis hint that the expression of Bag-1was correlated with poor prognosis. But there was no relationship with the expression of Bcl-2and Bax.Conclusion:1. The expression of Bag-1Bcl-2was higher in breast tumor tissues than in tumor-adjacent normal tissues, and was higher in IDC than in DCIS. The expression of Bag-1may be correlated with poor prognosis in IDC.2. The expression of Bag-1, Bcl-2and Bax indicate highly positive correlation in IDC and DCIS tumor tissues. Apoptosis-related genes Bag-1, Bcl-2and Bax may play important roles in the development of breast cancer.3. The expression of Bag-1was correlated with tumor size, but was not correlated with lymph node metastasis, clinical staging, expression of ER and PR in IDC. The expression of Bcl-2and Bax was not correlated with tumor size, lymph node metastasis, clinical staging, expression of ER and PR in IDC. |
