Objective To study the protective mechanism of the optimized composition of Danggui Saoyao Powder together with Mongolian medicine Chipaozi on model rats of chronic pelvic inflammation disease(CPID).To explore the regulatory effect of the optimized synergetic prescription on inflammatory-related cytokines and immune system in CPID model rats,to reveal the pathogenic mechanism of CPID at molecular level,and to provide a new idea for the research and development of CPID drugs.Methods The female SD rats were selected as the experimental object.CPID model was made by mixed bacteria combined with mechanical injury.The experimental rats were randomly divided into 7 groups: normal group,model group,Chipaozi group,Jingangteng capsule group,Danggui Shaoyao powder optimized synergetic prescription high,middle,low dose group.After 30 days of modeling,the drug was given continuously for 14 days.The serum IL-1?and IL-10,TNF-? in rats of each group were measured by enzyme linked immunosorbent assay(ELISA),and the histological changes of uterus in each group were observed under light microscope.Results Detection of serum by ELISA,compared with normal group,the content of IL-1 ? and TNF-? in model group increased and the content of IL-10 decreased significantly(P < 0.01).Compared with the model group,the optimized synergetic prescriptions could significantly increase IL-10,and decrease IL-1? and TNF-? in the middle and high dose groups(P < 0.01).The effect of Jingangteng capsule group on the increase of IL-10 and the decrease of IL-1?were significant(P < 0.01).There was no significant difference in the content of TNF-? between the Jingangteng capsule group and the model group(P > 0.05).The low dose and Chipaozi group had significant effect on the reduction of IL-1?and TNF-?levels(P < 0.01),there was no significant difference in the level of IL-10 between two groups above and the model group(P > 0.05).On the whole,the therapeutic effect of middle and high dose group on CPID rats was similar,which was superior to that of Jingangteng capsule group,and better than the low-dose group and the Chipaozi group.Under an optical microscope,compared with the normal group,the uterine cavity wall of the model group was not complete,the laminae propria was thin,the epithelial cells were partially shed and destroy,the necrosis and the inflammatory cell infiltration were involved in the whole layer of the uterine cavity,the uterine wall of the middle and high dose group and the Jingangteng capsule group remained intact,the epithelial cell hyperplasia was slight,and the infiltration of chronic inflammatory cells was less.Pathology score displayed: compared with model group,the high and middle dose group of the optimized Danggui Shaoyao Powder and the Jingangteng capsule group can effectively improve the above-mentioned pathological changes(P <0.05).Conclusion New Formula can produce the effective and positive effect on CPID rats.It provides some experimental and theoretical basis for the combined application of Chinese and Mongolian medicine in the treatment of CPID.It can regulate the immune response of the body by affecting the level of inflammatory factors.To reduce the possibility of adhesion by improving the local pathological microenvironment.It is suggested that the mechanism of its anti-chronic pelvic inflammation may be closely related to the increase of IL-10 content and the decrease of IL-1?and TNF-?content.
