| Bronchial asthma(referred to as asthma)is a common chronic airway inflammatory disease.In recent years,due to the changes of climate,environment,diet and other factors,the incidence,prevalence and mortality of asthma have been increasing year by year,and it has been listed as one of the “four stubborn diseases” by the World Health Organization.The pathogenesis of asthma is closely related to genetic,immune,environmental and other factors.At present,the immune imbalance of Th1/Th2 cells is regarded as an important pathogenesis of asthma.Pulvis Fellis Suis(PFS)is a traditional Chinese medicine used for the treatment of asthma.PFS has the effects of “clearing heat and moistening dryness,relieving cough and relieving asthma”,and is mainly used for the therapy of cough,asthma,throat obstruction and other diseases.The conjugated bile acid is the main component of PFS,which has good antiinflammatory and immunomodulatory activities.Based on this basis,the antitussive and expectorant effects of total conjugated cholic acid of PFS and its therapeutic effect on bronchial asthma were explored in this study.The antitussive and expectorant effects of total conjugated cholic acid were studied by concentrated ammonia water induced cough model and tracheal phenol red excretion method.The results showed that total conjugated cholic acid can significantly increase the cough latency,reduce the number of coughs,and increase the amount of tracheal phenol red excretion in mice,which suggested that total conjugated cholic acid has good antitussive and expectorant effects.On these basis,an ovalbumin(OVA)-induced mouse model of asthma was established to study the the therapeutic effect of total conjugated cholic acid on bronchial asthma.Lung tissue sections were stained by H&E and PAS for histopathological analysis.The number of total inflammatory cells and eosinophils in alveolar lavage fluid(BALF)were counted.IgE level in serum was determined by ELISA,and Th1 cytokine interferon-gamma(IFN-γ),Th2 cytokines interleukin(IL)-4,IL-5,IL-13,tumor necrosis factor alpha(TNF-α)and eotaxin in BALF and lung tissue were also determined by ELISA.The results showed that total conjugated cholic acid could alleviate the symptoms of asthma,reduce the degree of inflammatory cell infiltration in lung tissue,and inhibit excessive mucus secretion and goblet cell proliferation.Total conjugated cholic acid treatment could significantly reduce IgE level in serum,decrease Th2 cytokines(IL-4,IL-5 and IL-13)levels,as while as increase Th1cytokine(IFN-γ)level in BALF and lung tissue.In addition,total conjugated cholic acid treatment could also significantly reduce TNF-α and eotaxin levels in BALF and lung tissue.The above results indicate that total conjugated cholic acid of PFS has good antitussiveand expectorant effects and therapeutic effect on bronchial asthma.Total conjugated cholic acid can inhibit the airway inflammation and improve the pathological damage of lung tissue by regulating the imbalance of Th1/Th2 cell-associated pro-inflammatory cytokines and antiinflammatory cytokine,thereby inhibit the development of asthma.The above research conclusions suggested that total conjugated cholic acid of PFS could be a potential drug for the prevention and treatment of bronchial asthma. |
