Objective:To study the effect of Chuan Gu tablet on the expression of ERk1/2、p-JNK and ALP in rabbits with steroid-induced femoral head necrosis after modeling,and through light microscopy and electron microscopy The pathological changes of femur bone tissue specimens in rabbits were studied to investigate the efficacy and mechanism of Chuan Gu tablet in preventing and treating SANFH.Methods:Two groups were randomly separated from the 40 male rabbits of Japanese: the blank group and the model group.After 1 weeks of feeding,the model was established by the method of "He Xi Jing",with a period of 6 weeks.After 6 weeks,4 rabbits were chosen randomly in each group to identify whether the mold is successful.After modeling,the model groups were randomly divided into model control group,Chuan gu tablet treatment group and Xian ling gu bao capsule treatment group,with 8 rabbits in each group.The latter experiment was converted according to the 60 kg body weight conversion algorithm of adults.The treatment group was given gavage with medicine every day.The model control group was given distilled water every day,while the blank group was given no treatment for 42 days.After 42 days,collecting the abdominal aortic blood to calculate the serum ALP concentration of each group,and remove the right femoral head pathological sections.The tissue morphology of bone tissue was observed,and p-jnk and ERK protein contents were detected.Results:1、ALP:After modeling 6 weeks,compared with the model control group,the serum ALP content of the treatment group was reduced.However,there was no significant difference between Chuangu tablet group and Xianlinggubao group 2、p-JNK、ERK:After modeling 6 weeks,contrasted with the model matched group,the p-jnk gene expression in the treatment group was significantly down-regulated hile the comparison between the Chuangu tablet group and Xian ling gu bao group showed no statistical significance 3、ERK:After modeling 6 weeks,contrasted with the model matched group,the ERK gene expression in the treatment group was significantly up-regulated,while the difference between Chuangu tablet group and Xianlinggubao group has no statistically significant.Conclusion:1.The occurrence of SANFH was positively correlated with the expression of p-ERK1 and p-ERK2 proteins,and negatively correlated with the expression of p-ERK2 proteins.Chuangu tablet can up-regulate the expression of p-ERK1 and p-ERK2 related to proliferation in BMSCs MAPK signaling pathway of femoral head in SANFH model rabbits,down-regulate the expression of p-JNK related to apoptosis,promote the proliferation of BMSCs in femoral head and inhibit their apoptosis,so as to achieve the therapeutic effect of steroid-induced femoral head necrosis.2.Chuangu tablet can promote the expression of ALP in serum of SANFH model rabbits and promote bone growth.3.Chuangu tablet can effectively treat steroid-induced avascular necrosis of femoral head,and it has good curative effect and low price.It can reduce the economic and psychological burden of patients brought by the disease,as well as the social burden.
