Objective: This study was conducted to detect alteration of blood lipids and inflammatory cytokines IL-6,IL-8 and IL-10 after high-fat meal test in different fat acids tolerance groups to explore the association of lipid metabolism and inflammation.Methods: Volunteers who aged between 25 and 65 years old and signed an informed consent before the oral high-fat meal test(OFTT)and was eluted by normal diet for one week before OFTT.OFTT test was performed in the morning after 8-hour overnight fast.A special meal was finished within 10 minutes.Fasting blood and peripheral blood of 2h,4h and 6h after meal was collected to detect IL-6,IL-8,IL-10 and other indicators related to glucose and lipid metabolism.According to fasting and 4-hour postprandial TG level,participants were assigned into normal fat tolerance group(NFT,n=37),impaired fat tolerance group(IFT,n=36)and hypertriglyceridemia group(HTG,n=37).The separate definition of NFT,IFT and HTG was: fasting TG<1.7mmol/L and 4h TG<2.5mmol/L,fasting TG<1.7mmol/L and 4h TG≥2.5mmol/L,0h TG≥1.7mmol/L.All statistical analyses were performed by using SPSS 21.0 software.Results:1.No statistical difference of FBG and 2hPBG were found among the three groups(P>0.05).HTG group showed higher FINS and 2hINS levels than IFT group and NFT group(P<0.05).HOMA-IR in HTG group was significantly higher than that in NFT group(P<0.05).2.IFT group and HTG group showed significantly higher TC,LDL-C and ApoB than NFT group(P<0.01).The HDL-C in HTG group and IFT group was lower than that in NFT group(P<0.05).No significant difference was found in HTG vs IFT(P>0.05).The fasting ApoA1 showed no statistical difference among three groups(P>0.05).3.The three groups showed different time curves of TG level(F=120.871,P<0.01).The peak of TG level during OFTT was respectively observed to arise at 2h,4h and 6h in NFT,IFT and HTG group(P<0.01).TG in NFT group decreased slowly from 4h to 6h(P<0.01).The TG level at 6h in IFT group was still higher than fasting(P<0.01).Significant difference of plasma TG levels among the three groups was observed(F=70.911,P<0.01).Interaction between time and groups was significantly indicated(F=17.665,P<0.01).4.The level of IL-6 in three groups all gradually increased with time(F=32.858,P<0.01).The level of IL-6 in NFT group showed a peak at 4h after meal(P<0.01).The IL-6 level in IFT group increased gradually(P<0.05),and it was the most quickly at 2h(P<0.05).The IL-6 level in HTG group increased gradually after the meal(P<0.05),and the alteration was significant from 4h to 6h(P<0.01).Significant difference of plasma IL-6 levels among the three groups was observed(F=10.927,P<0.01).Interaction between time and groups was significantly indicated(F=4.255,P<0.01).5.The level of IL-8 in three groups all gradually increased with time(F=79.098,P<0.01).After meal,IL-8 level in NFT group was increased significantly at 2h(P<0.01),and decreased slightly at 6h(P<0.01).The IL-8 levels in IFT and HTG group were increased gradually after meal(P<0.01).There was a statistically significant difference of IL-8 among the groups(F=16.936,P<0.01).Interaction between time and groups was significantly indicated(F=4.482,P<0.05).6.The level of IL-10 in three groups all gradually decreased with time(F=50.553,P<0.01).IL-10 was firstly dropped in NFT group,and then began to rise at 6 hours(P<0.01).In IFT group,IL-10 decreased gradually after meal(P<0.01)and was basically stable from 4h to 6h(P<0.01).In HTG group,IL-10 decreased gradually after meal(P<0.01).There was a statistically significant difference of IL-10 among the groups(F=53.559,P<0.01).Interaction between time and groups was significantly indicated(F=6.827,P<0.01).Conclusion:1.With the impaired of fat tolerance disorder,the levels of pro-inflammatory factors such as IL-6 and IL-8 gradually increased and the anti-inflammatory factor IL-10 gradually decreased.2.This study suggested that postprandial TG metabolism in people with lipid metabolism disorder is slower than normal.
