Objective:Alcoholic Liver Disease(ALD)is a chronic liver disease caused by heavy drinking.The genetic mechanism plays an important role in its pathogenesis.Recently,the New England Journal found that the HSD17B13 rs72613567 allele alleviated the risk of liver damage caused by the genetic susceptibility of PNPLA3 P.I148M variants.Due to the ethnic differences between ALD patients,it is necessary to verify in Chinese Han population.The aim of this study was to investigate the genetic susceptibility of Single-Nucleotide Polymorphisms of gene polymorphism candidate genes HSD17B13,ALDH2,and OAS1 to ALD patients in Chinese Han population.Methods:This study was a multicenter large sample study involving 769 ALD patients and767 healthy controls using Matrix-Assisted Laser Desorption/Ionization Time of Flight Mass Spectroscopy(MALDI-TOF MS).Methods Five genotyping sites of the following genes were genotyped:HSD17B13(rs72613567,rs117111943,rs6834314),ALDH2 rs671,OAS1 rs11066453,correlation analysis using PLINK 1.07 software.Results:The alleles and genotypes of rs6834314 of HSD17B13 gene were statistically different between the two groups(P=8.2×10-3;P=0.02).Statistics were obtained under additive model,dominant model and recessive model.The difference was(P=8×10-3;P=0.04;P=0.02);the allele of rs117111943 was statistically significant(P=9×10-3),and its genotype was in two.There was no statistical difference between the groups,and there was statistical difference in the additive model(P=6×10-3).There was no statistical difference in the dominant model and the recessive model;the allele of the SNP locus rs72613567 There were significant differences between the disease group and the control group(P=0.01,P=0.04).There were statistical differences between the two groups in the additive model and the dominant model(P=0.01;P=0.03),there was no statistical difference under the recessive model(P=0.06);the difference between the ALDH2 rs671 allele in the disease group and the control group was statistically significant(P=4.18×10-43),There was no statistical difference between the two groups in the genotype,and there were statistical differences in the additive model(P=1.21×10-42).There was no statistical difference between the dominant model and the recessive model.The difference in the allele of OAS1 rs11066453between the disease group and the control group was statistically significant(P=5.70×10-12),and the genotype was not statistically different between the two groups.Statistics were obtained under the additive model.Learning differences(P=4.74×10-18),there is no statistical difference between the two groups in the dominant model and the recessive model.Conclusion:This study found that HSD17B13 gene polymorphisms rs6834314 and rs72613567are significantly associated with ALD in Chinese Han population.ALDH2 rs671 is potentially relevant in the Chinese Han ALD population.OAS1 rs11066453 and HSD17B13 rs117111943 have no obvious susceptibility in Chinese Han population ALD.
