| Objective: In this study,the expression of SOCS3,E-cadherin and N-cadherin in 50 cases of clear cell renal cell carcinoma(CCRCC)and its adjacent tissues were detected by immunohistochemistry(Envision method)at protein level,and their roles in the clear cell renal cell carcinoma were analyzed.Methods:(1)To collect the data of patients with renal cell carcinoma who were diagnosed as clear cell renal cell carcinoma by histopathology from January 2008 to January 2018 in Urology Department of the Central Hospital of EnShi Tujia and Miao Autonomous Prefecture,including patient’s sex,age,telephone number,date of operation,date of diagnosis,pathology,clinical staging and grade.The patients were followed up by telephone and concerned the survival of patients.(2)The paraffin-embedded tissues of clear cell renal cell carcinoma and its adjacent tissues were colleted from the pathological Department of the Central Hospital of EnShi Tujia and Miao Autonomous Prefecture.The expression levels of SOCS3,E-cadherin and N-cadherin in clear cell renal cell carcinoma and its adjacent tissues were detected by immunohistochemistry(Envision method).(3)SPSS17.0 statistical software was used for statistical analysis.The difference of SOCS3,E-cadherin and N-cadherin expression in clear cell renal cell carcinoma tissues and adjacent tissues was tested by chi-square test.The relationship between the expression of SOCS3,E-cadherin and N-cadherin in renal clear cell carcinoma and pathological grade,clinical staging and lymph node metastasis of patients was also tested by chi-square test.The correlation between SOCS3 and E-cadherin or N-cadherin was analyzed by spearman grade correlation.The survival time and survival curve were analyzed by Kaplan-Meier method.P < 0.05 was statistically significant.Results:(1)The positive expression rate of SOCS3 in the clear cell renal cell carcinoma was 32.00%(16/50)and 86.00%(34/50)in adjacent tissues,respectively.The positive expression rate of E-cadherin was 40.00%(20/50),while the E-cadherin was expressed in all the adjacent tissues.The positive expression rate of N-cadherin in the clear cell renal cell carcinoma was 64.00%(32/50)and 2.00%(1/50)in adjacent tissues,respectively.(2)The expression of SOCS3 and E-cadherin in clear cell renal cell carcinoma is not correlated with patient gender and age,but closely related to pathology grade,clinical stage and lymph node metastasis(P< 0.05).With the increase of pathology grade and clinical stages,the positive expression rate of the SOCS3 and E-cadherin decreased.But SOCS3 and E-cadherin expression was lower in patients with lymph node metastasis.The expression of N-cadherin protein was not related to the gender,age,pathology grade and clinical stage of patients with clear cell renal cell carcinoma,but related to the lymph node metastasis.High expression of N-cadherin was found in patients with lymph node metastasis,while low expression of N-cadherin was found in patients without lymph node metastasis(P<0.05).(3)Correlation analysis results showed that the expression of SOCS3 and E-cadherin in clear cell renal cell carcinoma was positively correlated,while the expression of SOCS3 and N-cadherin was negatively correlated,with correlation coefficients of 0.452 and-0.348,respectively(P <0.05).The difference was statistically significant.(4)Patients were followed up for 0-120 months,and the median survival time was 71.00±8.96 months.60.00%(30/50)of patients with clear cell renal cell carcinoma survived,and 36.00%(18/50)of them died,of which 32.00%(16/50)died of recurrence or metastasis of clear cell renal cell carcinoma,4.00%(2/50)died of other diseases,and 4%(2/50)lost follow-up.The survival analysis suggested that the low expression of SOCS3 and E-cadherin and high expression of N-cadherin were correlated with the survival time of patients with clear cell renal cell carcinoma.Conclusion:(1)The expression of SOCS3 and E-cadherin protein is negatively correlated with clinical stage,grade and lymph node metastasis of clear cell renal cell carcinoma,However,the expression of N-cadherin protein was only positively correlated with lymph node metastasis in patients with clear cell renal cell carcinoma.(2)SOCS3 can inhibit the epithelial mesenchymal transformation(EMT)of clear cell renal cell carcinoma,and further inhibit the local metastasis and invasion of renal cell carcinoma. |
