BackgroundRenal cell carcinoma is a relatively common malignant cancer of the urinary system,and its incidence ranks second only to bladder cancer in malignant cancer of human urinary system.Recently,more and more studies have shown that mi R-221,a member of the micro RNA family,has elevated expression levels in different malignant tumor tissues.Epithelial-mesenchymal transition,which converts polarity-fixed epithelial cells into mesenchymal cells,is often accompanied by a decrease in the expression of the epithelial marker E-cadherin and an increase in the expression of the mesenchymal marker N-cadherin.In this study,the expression of mi R-221 and the expression of E-cadherin,N-cadherin in renal clear cell carcinoma were detected and their correlations were analyzed.Then further explore the relationship between the expression level of mi R-221 and the survival and prognosis of patients with renal clear cell carcinoma.ObjectiveTo study the expression level of mi R-221 in renal clear cell carcinoma,and analyze the relationship between the expression of mi R-221 and various pathological parameters.At the same time,the expressions of E-cadherin and N-cadherin in renal clear cell carcinoma were detected,and the correlation between the expression of mi R-221 and E-cadherin and N-cadherin was analyzed.Finally,we explored the relationship between the expression of mi R-221 and the survival time and prognosis of renal cell cancer patients.Methods1.Collect 38 cases of cancer tissue specimens diagnosed as clear cell renal cell carcinoma by pathology as the experimental group,and collect the normal tissue specimens adjacent to the cancer as the control group,and use quantitative real-time PCR to detect the expression of mi R-221 in the two groups respectively.Then,the relationship between mi R-221 and pathological characteristics of renal clear cell carcinoma patients was analyzed.2.Immunohistochemical staining was performed on the pathological sections of cancer tissues and adjacent tissues to explore the expression of E-cadherin and N-cadherin in cancer tissues and corresponding adjacent tissues.3.Using Spearman correlation analysis to analyze the correlation between the expression of E-cadherin and N-cadherin and the expression of mi R-221 in renal clear cell carcinoma.4.To analyze the relationship between E-cadherin,N-cadherin and clinicopathological characteristics of patients with renal clear cell carcinoma.5.Kaplan-Meior survival analysis and Log-rank test were used to evaluate the relationship between the expression level of mi R-221 in cancer tissues of patients with clear cell renal cell carcinoma and the prognosis of patients’ survival.Results1.The results of q RT-PCR showed that the expression of mi R-221 in the cancer tissue group(4.911±1.064)was significantly higher than that in the control adjacent tissue group(1.163±0.238),and the difference was statistically significant(P<0.05).In cancer tissue specimens of patients with renal clear cell carcinoma,the relative expression of mi R-221 was related to pathological grade and lymph node metastasis(P<0.05),but not to age,gender,tumor size,and tumor thrombus in renal vein.2.The results of immunohistochemistry showed that the expression positive rate of E-cadherin in the cancer tissue was 26.3%(10/38),and the expression positive rate in the control adjacent tissue group was 81.6%(31/38).The expression positive rate of N-cadherin in the cancer tissue was 68.4%(26/38),and the expression positive rate in the control adjacent tissue group was 7.9%(3/38).There was a statistically significant difference between the two groups(P<0.05).3.The results of Spearman correlation analysis showed that the expression of E-cadherin in renal clear cell carcinoma was negatively correlated with the expression of mi R-221,the correlation coefficient was r =-0.569,P<0.001.The expression of N-cadherin was positively correlated with the expression of mi R-221,the correlation coefficient was r =0.660,P<0.001.4.The expression of E-cadherin in clear cell renal cell carcinoma has nothing to do with age,gender,tumor size,lymph node metastasis,tumor thrombus formation,but is related to pathological grade.The expression of N-cadherin was not related to age,gender,tumor size,tumor thrombus formation,lymph node metastasis and pathological grade.5.The Kaplan-Meior survival curve was used to study the relationship between the expression of mi R-221 and the survival time of patients with clear cell renal cell carcinoma.The results showed that the 5-year survival of the mi R-221 high expression group was52.9%(9/17),and the 5-year survival of the mi R-221 low expression group was 80.9%(17/21).Log-rank test evaluated the difference in survival curve between the mi R-221 high expression group and the mi R-221 low expression group,and the results showed that the difference was statistically significant(P<0.05).ConclusionThis study shows that mi R-221 may plays a role in the progression of EMT in clear cell renal cell carcinoma,and is a potential marker for survival and prognosis assessment of clear cell renal cell carcinoma patients.
