Objective:In this study,we observed the effect of warming needling at Yaoyangguan and Xiliao points on serum cortisol(Cor),histamine(HIS),intrauterine HIS and its H1 receptor protein(H1R)in primary dysmenorrhea model rats,and explored its mechanism,so as to provide experimental and theoretical basis for the treatment of primary dysmenorrhea by warming needling and moxibustion.Methods:Seventy-two rats were randomly divided into 6 groups,including pre-meridian warming acupuncture group,post-meridian warming acupuncture group,pre-meridian warming acupuncture group,post-meridian western medicine group,model group and blank group,with 12 rats in each group.The primary dysmenorrhea model of rats was established by the combination of oxytocin and estradiol benzoate.The warm needling group was treated with warm needling at Yaoyangguan and Xiliao points,and the western medicine group was treated with ibuprofen by gastric lavage.Among them,the warming needling group started on the 7th day after injecting estradiol benzoate(E2)into the femoral subcutaneous area,and the warming needling group started on the 11th day after injecting E2 into the femoral subcutaneous area,and the warming needling group was the same as the warming needling group before the menstrual area.From the 7th day after E2 was injected subcutaneously into the femur,ibuprofen was given orally 20 mg/kg per day in the former western medicine group,and from the 11th day after E2 was injected subcutaneously into the femur in the latter western medicine group,the same drug was given orally as the former western medicine group.The latency of writhing reaction and the number of writhing in 30 minutes were counted after5 consecutive days.Anatomical samples were taken and the levels of Cor,HIS and HIS in serum and uterus were measured by ELISA.The levels of H1R were measured by Western blot(Wb).Around the changes of writhing reaction,uterine histamine and serum cortisol in six groups before and after treatment,the therapeutic effects of blank group,model group,pre-and post-menstrual warm needling group,pre-and post-menstrual western medicine group were compared.Results:(1)Twist response:There was no twist in the blank group,and the number of twists in the model group was more and the latency of twists was longer observed.There was a significant difference between the two groups(P<0.01).Compared with the model group,The incubation period of the first torsion was significantly prolonged in the pre-warming acupuncture group,the post-warming acupuncture group and the pre-warming western medicine group,and the number of torsion reactions within 30 minutes was reduced(P<0.01).There was no significant difference in latency and frequency of torsion between model group and postmenopausal western medicine group(P>0.05).There was no significant difference in latency of torsion between pre-warming acupuncture group,post-warming acupuncture group and pre-warming western medicine group(P>0.05).(2)Serum Cor and HIS content:Compared with blank group,the content of HIS and Cor in model group,pre-warming acupuncture group,post-warming acupuncture group,pre-warming western medicine group and post-warming western medicine group increased significantly(P<0.01).Compared with the model group and the post-menstrual western medicine group,the contents of HIS and Cor in the pre-menstrual warm needling group,the post-menstrual warm needling group and the pre-menstrual western medicine group decreased significantly(P<0.01),and the differences between the model group and the post-menstrual western medicine group were significant(P<0.05).There was no significant difference in the content of HIS and Cor between pre-warming acupuncture group,post-warming acupuncture group and pre-warming western medicine group(P>0.05).(3)Uterine HIS and its H1R level:Compared with the blank group,the HIS content and H1R level in the model group,the pre-warming acupuncture group,the post-warming acupuncture group,the pre-warming acupuncture group and the post-warming western medicine group increased significantly,and the H1R level decreased significantly(P<0.01).Compared with model group and postmenopausal western medicine group,the content of HIS decreased significantly and the expression of H1R increased significantly in pre-warming acupuncture group,post-warming acupuncture group and pre-menopausal western medicine group(P<0.01).There was no significant difference in HIS level among pre-warming acupuncture group,post-warming acupuncture group and pre-warming western medicine group(P>0.05),but there was significant difference in H1R level(P<0.05).Conclusion:(1)Warm acupuncture and moxibustion may play an analgesic role by reducing the content of serum HIS and Cor,reducing the level of HIS in uterine tissue and increasing the level of H1R in uterus.(2)Warm acupuncture and moxibustion have strong anti-inflammatory and analgesic effects on dysmenorrhea of PD model rats,and effectively improve the quality of life of PD model rats.That is to say,warming acupuncture and moxibustion has a better preventive and therapeutic effect on primary dysmenorrhea.(3)Warm needle moxibustion has remarkable curative effect on PD, and has certain advantages and development potential,which can be popularized and applied in clinic. |
