Objective To explore the serum level and clinical significance of β-catenin,DKK-1and MMP-9 in the knee osteoarthritis(KOA)combined with osteoporosis(OP)patients.Methods ELISA was used to test the serum level of β-catenin、DKK-1 and MMP-9in 60 KOA patients combined with OP patients(OA + OP group),60 KOA patients(OA group),60 OP patients(OP group)and 35 healthy volunteers(HC group).The serum level of β-catenin、DKK-1 and MMP-9 were compared in the four groups.The correlation between serum β-catenin、DKK-1 and MMP-9 with lumbar spine BMD、femoral neck BMD 、 lumbar spine T-score 、 lumbar spine Z-score 、 femoral neck T-score、femoral neck Z-score in OA+OP group was analyzed.The correlation between serum β-catenin,DKK-1 and MMP-9 was analyzed in OA+OP group was analyzed.Results 1.The level of serum β-catenin in OA + OP group(34.70 ±21.95pg/m L)lower than HC group(47.97 ± 16.93 pg/m L),lower than OA group(54.58 ±13.43pg/m L),higher than OP group(29.96 ± 14.31 pg/m L),differences were statistically significant(P < 0.05);OP group lower than HC group and the difference was statistically significant(P < 0.05);OA group is higher than HC group but there was no statistically significant difference(P > 0.05).2.The serum level of DKK-1 in OA + OP group(5.82 ± 1.78ng/m L)is higher than HC group(5.00± 1.47ng/m L),higher than OA group(3.97 ±1.39ng/m L),difference had statistical significance(P < 0.05).OA + OP group is lower than the OP group(6.55± 0.52 ng/m L),but there was no statistically significant difference(P > 0.05);OA group is lower than HC group,the difference was statistically significant(P < 0.05);The OP group is higher than HC group difference was statistically significant(P <0.05).3.The serum level of MMP-9 in OA + OP group(709.50 ±382.83ng/m L)is higher than HC group(270.40 ± 119.24ng/m L),higher than OA group(452.33 ±270.81ng/m L),higher than OP group(413.23 ± 186.03 ng/m L)and the differences were statistically significant(P < 0.05).Although the OA and OP group were higher than that of HC group,but there was no statistically significant difference(P > 0.05).4.The serum level of β-catenin in OA + OP group was positively correlate with lumbar spine BMD(r = 0.479,p < 0.031)、femoral neck BMD(r = 0.64,p < 0.0001)、lumbar spine T-score(r=0.6691,P<0.0001)、lumbar spine Z-score(r=0.7353,P<0.0001)、femoral neck T-score(r=0.6608,P<0.0001)、 femoral neck Z-score(r=0.7026,P<0.0001).The serum level of DKK-1 in OA + OP group was negatively correlate with lumbar spine BMD(r=-0.454,p=0.037),femoral neck BMD(r=-0.215,p= 0.0013),lumbar spine T-score(r=-0.709,P<0.0001)、lumbar spine Z-score(r=-0.6884,P<0.0001)、femoral neck T-score(r=-0.8392,P<0.0001)、femoral neck Z-score(r=-0.7319,P<0.0001).The serum level of MMP-9 in OA + OP group was negatively correlate with lumbar spine BMD(r=-0.631,p<0.0001),femoral neck BMD(r=-0.392,p=0.035),lumbar spine T-score(r=-0.7059,P<0.0001)、lumbar spine Z-score(r=-0.7379,P<0.0001)、femoral neck T-score(r=-0.5241,P<0.0001)、femoral neck Z-score(r=-0.7319,P<0.0001).5.In OA+OP group,the level of β-catenin was negatively correlate with DKK-1(r=-0.5533,p<0.0001),the serum level of β-catenin was positively correlate with MMP-9(r=0.5586,p<0.0001).No significant association is found between DKK-1 and MMP-9(r=0.0265,p=0.2854).Conclusion 1、Wnt/ β-catenin signaling pathway and its related regulatory factors may be involved in the pathogenesis of KOA combined with OP;2、The joint detection of β-catenin、DKK-1 and MMP-9 is of great value in judging the bone metabolic status of KOA combined with OP.
