Objective:To screen the prescriptions with a significant inhibitory effect on the proliferation of human esophageal carcinoma cells of TE-1,EC-1,EC109 and EC9706 from the 111 prescriptions of Treatise on Febrile Diseases.To explore the mechanism of Chinese medicine on the treatment of esophageal carcinoma through observing the effect of prescription on biological characteristics,with which provide basis and reference for clinical treatment and experimental study of esophageal carcinoma.Methods:1.Taking esophageal carcinoma cells TE-1,EC-1,EC109 and EC9706 as models,use MTT method to select effective prescriptions which could effectively inhibit cell proliferation from the 111 prescriptions of Treatise on Febrile Diseases,further select the most effective prescription.2.Observe and record the morphological changes of cells by inverted microscope.3.Explore the influence of screened prescriptions on the cycle of TE-1,EC-1,EC109 and EC9706 cells by using the flow cytometry.4.Use RTCA DP analyzer to analyze the effect of the screened prescriptions on the proliferation,migration and infiltration of TE-1,EC-1,EC 109 and EC9706 cells.5.The effect of the selected formula on clonogenesis ability of TE-1,EC-1,EC 109 and EC9706 cells were tested by Soft Agar Assay.Results:1.Having Screened 4 prescriptions with a significant inhibitory effect on the proliferation of human esophageal carcinoma cells of TE-1,EC-1,EC 109 and EC9706 from the 111 prescriptions of Treatise on Febrile Diseases,Baitouweng Decoction,Dahuanghuang-lianxiexin Decoction,Xiaoxianxiong decoction and Guadi powder,respectively.2.The effect of 4 prescriptions on the proliferation of human esophageal carcinoma cells increase with the increase of drug concentration and time extention.3.Four prescriptions can make a significant difference in cell morphology.4.The cell cycles of TE-1,EC-1,EC109 and 9706 are all affected by Baitou weng Decoction,Dahuanghuanglianxiexin Decoction,Xiaoxianxiong decoction and Guadi powder.Compared with the control group,the number of G1 phase cell about TE-1 and EC 109 incrased in the group of Baitouweng Decoction,Dahuang-huanglianxiexin Decoction and Xiaoxianxiong decoction.While the number of G1 phase cells decreased and the S phase cells increased about EC-1 cells in the group of Baitouweng Decoction,Dahuanghuanglianxiexin Decoction and Xiaoxianxiong decoction.The number of G1 phase cells decreased and the S phase cells increased obviously about EC9706 cells in the group of Dahuanghuanglian xiexin Decoction and Xiaoxianxiong decoction,meanwhile the G2 phase cells increaseed obviously in the group of Baitouweng Decoction.The number of G1 phase cells decreased and the G2 phase cells increased about TE-1,EC-1,EC 109 and 9706 cells in the group of Guadi powder.5.Baitouweng Decoction,Dahuanghuanglianxiexin Decoction,Xiaoxianxiong decoction and Guadi powder all can inhibit the proliferation,migration and infiltration of TE-1,EC-1,EC109 and EC9706 cells.6.Clonogenesis ability of TE-1,EC-1,EC109 and EC9706 cells can be inhibited by Baitouweng Decoction,Dahuanghuanglianxiexin Decoction,Xiaoxianxiong decoction and Guadi powder.Conclusions:1.Baitouweng Decoction,Dahuanghuanglianxiexin Decoction,Xiaoxianxiong decoction and Guadi powder have a significant inhibitory effect on the proliferation of human esophageal carcinoma cells of TE-1,EC-1,EC109 and EC9706.2.There is a dose-dependent and time-dependent relationship between the efficacy and Baitouweng Decoction,Dahuanghuanglianxiexin Decoction,Xiaoxianxiong decoction and Guadi powder.3.Baitouweng Decoction,Dahuanghuanglianxiexin Decoction,Xiaoxianxiong decoction and Guadi powder have different blocking effects on cell morphology.4.The cell cycles of TE-1,EC-1,EC109 and EC9706 are all affected by Baitou-weng Decoction,Dahuanghuanglianxiexin Decoction,Xiaoxianxiong decoction and Guadi powder.5.Baitouweng Decoction,Dahuanghuanglianxiexin Decoction,Xiaoxianxiong decoction and Guadi powder all can inhibit the proliferation,migration and infiltration of TE-1,EC-1,EC 109 and EC9706 cells.6.Clonogenesis ability of TE-1,EC-1,EC109 and EC9706 cells can be inhibited by Baitouweng Decoction,Dahuanghuanglianxiexin Decoction,Xiaoxianxiong decoction and Guadi powder. |
