The Relationship Between Oral Lichen Planus And NF-κB Pathway Correlation Factors And The Therapeutic Effect Of Prescription Huashi Xingyu Qingre Decoction On Oral Lichen Planus

Part one Relationship between oral lichen planus and NF-κB pathway correlation factorsObjective:To investigate the expression of NF-κB(nuclear factor-κB)signaling pathway-related cytokine IL-8(Interleukin-8),RANTES(Regulated upon activation normal T cell expressed and secreted facto)in patients with OLP and healthy controls,and to analyze the correlation of the expression of IL-8 and RANTES,then to explore the role of NF-κB signal transduction system in the occurrence and development of oral lichen planus.Methods:1 We selected 30 patients with oral lichen planus from December 2015 to June 2017 from the Fourth Hospital of Hebei Medical University as experimental group.Partial pathological mucosal tissue of patients with pathological resection before treatment.In the control group,30 healthy patients were selected from the stomatology Department of the 4th hospital of Hebei Medical University for the removal of impacted teeth,They were both ambushed and never had wisdom tooth periodontitis.2 The expression of NF-κB related factor IL-8 and RANTES in the patients with OLP and control group were detected by RT-PCR(Reverse Transcription-Polymerase Chain Reaction).Analysis of the expression differences between the patients with OLP and the healthy control group and the correlation of IL-8 and RANTES expression.Results:1.RT-PCR results showed that IL-8 was expressed in both patients with OLP and the healthy control group,however,the expression of OLP was significantly higher than that of normal control group.(t=-12.23,P<0.05).2.RANTES were expressed in both patients with OLP and healthy controls,but the expression of RANTES in patients with OLP was significantly higher than that of normal controls.(t=11.615,P<0.05).3.The expression of IL-8 and RANTES was significantly correlated in the patients with OLP.(R=0.658,P<0.05).Part two The therapeutic effect of prescription HuashiXingyu Qingre decoction on oral lichen planusObjective:The results of the previous study in this group showed that the expression levels of IL-8 and RANTES in saliva and serum of OLP patients were higher than that of normal people,and in order to further study and confirm the obvious curative effect and the possible therapeutic mechanism of treating OLP in prescription Huashi Xingyu Qingre decoction,this study used ELISA(enzyme-linked immunosorbent assay)method to analyze the expression of IL-8 and RANTES in the saliva and serum of OLP patients after treated with the HuashiXingyu Qingre decoction,to provide theoretical basis for the clinical application of this prescription.Methods: The experimental group is 30 patients with OLP in the first Part,collected the venous blood and saliva before and after treatment.The interval was 8 weeks.The blood is centrifuged to get only the supernatant.Saliva serum is divided into sterile EP tubes and stored at-80℃refrigerator.The expression of IL-8 and RANTES in serum and saliva of patients with OLP were quantitatively studied by ELISA.Results:ELISA results showed that the serum and saliva concentrations of IL-8 and RANTES decreased significantly after treatment with OLP.(P<0.05).Conclusions:1.In the tissue of OLP patients,the related factors IL-8,RANTES cytokines were higher than those of control group to varying degrees.It is assumed that the occurrence and development of oral lichen planus may berelated to the inflammation pathway guided by NF-κB and its mediated inflammatory factors.2.The expression of IL-8 and RANTES in the tissue of OLP patients was significantly related,suggesting that IL-8 and RANTES may have synergistic pathogenic effects in the pathogenesis of OLP.3.Treatment of oral lichen planus with prescription HuashiXingyu Qingre decoction has obvious clinical effect.4.The expression levels of saliva and serum IL-8 and RANTES in OLP patients were significantly reduced after the treatment of HuashiXingyu Qingre decoction.It was suggested that the HuashiXingyu Qingre decoction may be used to treat OLP by inhibiting the release of related inflammatory factors in the NF-κB signaling pathway.
