Part one The expression of CXCL12,CXCR4,MMP-9mRNA in Oral Lichen Planus tissueObjective: Oral lichen planus(oral lichen planus OLP)is a chronic inflammatory disease with unknown etiology.Most scholars believe that immune factors play a leading role in.The study of its immune pathogenesis is of great importance for the treatment of OLP.In recent years,chemokine CXCL12 and its receptor CXCR4 have been involved in the immunological pathogenesis of various inflammatory diseases became study hot point.As a protein factor that can be secreted by many immune cells,MMP-9 participates in the inflammatory response,and studies have shown that MMP-9 is involved in the pathogenesis of OLP.The CXCL12/CXCR4 biological axis can induce MMP-9 to secrete through the nuclear factor-kappa B(nuclear factor-KB NF-ΚB)and other related pathways.The previous study of OLP was closely related to the NF-ΚB pathway and its related cytokines.Whether the biological axis of the CXCL12/CXCR4 is involved in the pathogenesis of OLP and whether the axis can also pass through some related pathway to effect the secretion of MMP-9 on OLP needs further study.In this study,Real-time quantitative fluorescence PCR(Quantitive Real-time PCR qPCR)was used to detect expression of CXCL12,CXCR4,MMP-9mRNA in OLP tissues,to analyse the relativity of CXCL12,CXCR4,MMP-9 to explore the effect of CXCL12/CXCR4 axis and MMP-9 on the pathogenesis of OLP.Methods : QPCR experimental technique was used to determine the expression of CXCL12,CXCR4,MMP-9mRNA in OLP tissue(Erosive and non-erosive)and Normal mucous tissue,preliminarily analysed the expressiondifference of CXCL12,CXCR4,MMP-9mRNA in different types of OLP and normal mucosa.The mechanism of CXCL12/CXCR4 axis on OLP and its relationship with MMP-9 in the pathogenesis of OLP were discussed.Results:1.QPCR results show CXCL12,CXCR4 and MMP-9mRNA were expressed in OLP tissues and normal mucosa tissues.The expression of the three factors in OLP tissues was significantly higher than that in normal mucosal tissues,and the difference was statistically significant(P <0.05).2.OLP typing(Erosive and non-erosive)Intratissue,There was no significant difference in CXCL12,CXCR4,MMP-9mRNA expression(P>0.05).3.Pearson correlation analysis showed that there is a positive correlation between CXCL12,CXCR4 and MMP-9.(r=0.509,P=0.001;r=0.530,P=0.000).Part two The effection of Huashixingyu Qingre Recipe on expression of CXCL12,CXCR 4,MMP-9 in oral lichen planusObjective : The complex etiology of OLP increases the difficulty of clinical diagnosis and treatment,and no special drugs and methods have been found.Previous studies found that Huashixingyu Qingre recipe has a significant effect on OLP,and it may regulate many cytokines and signal pathways involved in the pathogenesis of OLP by analysis.This experiment was used to study the expression of CXCL12,CXCR4 and MMP-9 in serum with OLP before and after the intervention of Huashixingyu Qingre recipe.The expression of CXCR4 and MMP-9 were studied to further explore the mechanism of Huashixingyu Qingre in the treatment of OLP,to further evaluate its clinical effect,and to lay a theoretical foundation for clinical treatment of OLP.Methods: Enzyme-linked immunosorbent assay(ELISA)was used to determine the level of CXCL12,CXCR4,MMP-9 in Serum of 25 normal persons as contrast,to determine the expression variety of CXCL12,CXCR4,MMP-9 in serum of 32 patients with OLP(Erosive and non-erosive)wasmeasured before and after taking Huashixingyu Qingre prescription.And the clinical records were used to analyze the therapeutic effect of the prescription.Results:1.CXCL12,CXCR4,MMP-9 were expressed in OLP patients with different typing(Erosive and non-erosive)and normal persons.The expression of the former was higher than that of the latter,and the difference was statistically significant(P<0.05).There was no significant difference in the expression of CXCL12,CXCR4 and MMP-9 between Erosive OLP and Non-erosive OLP serum and the difference was not statistically significant(P>0.05).2.The expression of CXCL12,CXCR4,MMP-9 in serum of OLP was significantly after treatment with the prescription of removing dampenss and removing blood stasis and clearing away heat lower than that before treatment,and the difference was statistically significant.(P<0.05).3.After Huashixingyu Qingre recipe(8Weeks):17cases showed remarkable effect(53.13%);effective 12 cases(37.5%);invalid 3 cases.The total efficiency was 90.63%.The clinical effect of Huashixingyu Qingre recipe on OLP is relatively good.Conclusions:1.The expression of CXCL12,CXCR4 and MMP-9mRNA in OLP was significantly higher than that in normal mucosa.There was no significant difference in the expression of CXCL12,CXCR4 and MMP-9 between Erosive OLP and Non-erosive OLP.The three factors may be involved in the pathogenesis of OLP.2.The expression of gene level CXCL12,CXCR4 and MMP-9 in OLP tissues was positively correlated,suggesting that CXCL12/CXCR4 and MMP-9 may play a synergistic role in the pathogenesis of OLP.3.The expression of CXCL12,CXCR4,MMP-9 in OLP serum was h-igher than that in normal serum,further proof of CXCL12,CXCR4,MMP-9 may be involved in the pathogenesis of OLP.4.In serum of OLP after treatment with Huashixingyu Qingre prescription,the expression of CXCL12,CXCR4,MMP-9 was lower than that before treatment,which suggested that the prescription after removing da-mpness and stasis might influence the expression of CXCL12,CXCR4,MMP-9 plays a role in the treatment of OLP.5.The clinical effect of Huashixingyu Qingre recipe on OLP was r-emarkable. |
