| Objective:To explore the effects of Pokemon silence on C-myc expression in human breast cancer cells(T47D and MCF-7)and to analyze the clinical correlationship between Pokemon and C-myc.Methods: Experiment was divided into three parts.Part I: T47 D,MCF-7 breast cancer cells cultured in vitro were selected as cell models.The si RNA silencing the Pokemon specifically and the Scrambled si RNA were transiently transfected to the above two breast cancer cells.Western blot and q RT-PCR experiments were used to analyze the silencing effect of Pokemon-si RNA on Pokemon.Then we used western blot and q RT-PCR experiments respectively to detect the effects of Pokemon-silencing on the m RNA expression levels and the protein expression levels of C-myc.Part II: Building the C-myc promoter,which was co-transfected to T47 D and MCF-7 cells with the Pokemon-si RNA and the Scrambled si RNA respectively.The C-myc promoter activity was detected by the Dual-Luciferase reporter assay.Part III: Both the tissue microarray with 100 cases of human breast invasive ductal carcinoma tissues and 10 cases of adjacent normal breast tissues and the immunohistochemistry staining method were used to reveal the differential expression of Pokemon and C-myc.Results: 1)Pokemon influences the expression of C-myc in breast cancer cells: The low-expression of Pokemon can reduce the expression levels of m RNA and protein of C-myc in T47D、MCF-7 breast cancer cells.2)The mechanism of Pokemon on C-myc in breast cancer: The low-expression of Pokemon can reduce the activity of C-myc promoter in T47D、MCF-7 cells.3)The clinical correlationship between Pokemon and C-myc: By the immuno-histochemistry staining method for 100 cases of human breast invasive ductal carcinoma tissues,we can know that the expression of Pokemon and C-myc was positively correlated relationship,the difference was statistically significant(R=0.464,p=0.000).Conclusion:1)Silencing Pokemon can inhibit the expression of C-myc in breast cancer cells,which might be related to inhibiting the activity of C-myc promoter.2)The expression of Pokemon and C-myc was positively correlated relationship in breast cancer tissues. |
