| Objective: To study the effect of Pokemon on ER、PR expression in human breast cancer cells and to analyze the clinical correlationship between Pokemon and ER、PR.Methods: The experiment is divided into three parts.First,In vitro cultured breast cancer MCF7 and T47 D cells as the research object,designed pokemon-specific siRNA,transient transfected siRNA to MCF7 and T47 D cells,We analyzed the silencing effect of siRNA on pokemon and constructed the MCF7 T47 D cell which silenced Pokemon.The effect of pokemon on the expression of ER and PR protein was detected by Western Blot and the nucleic acid level was detected by q-RTPCR;The promoter activity was detected in MCF7 and T47 D cells by constructing ER promoter sequence.The ER promoter plasmids and siRNA were transiently co-transfected into MCF7 cells.The effect of Pokemon on ER promoter activity was analyzed by luciferase reporter assay,and the mechanism of Pokemon on ER expression was analyzed.Last,Immunohistochemical method was used to detect the expression of Pokemon,ER,and PR in 100 cases of non-specific invasive ductal carcinoma of the breast,10 cases of marginal tissue,and 50 cases ofdifferent pathological types of human breast cancer and 50 cases of metastatic lymph nodes with different pathological types,analysis of the three in clinical relevance.Results 1).q-RTPCR and Western Blot analysis showed that Pokemon had an effect on the expression of ER and PR in breast cancer cells(MCF7and T47D).silencing Pokemon expression on the expression of ER,PR nucleic acid and protein.2).Luciferase reporter gene activity experiments showed that silencing Pokemon can inhibit the activity of ER promoter in MCF7 and T47 D cells;3).Immunohistochemical analysis 100 cases of human non-specific invasive ductal carcinoma of the breast and 10 cases of marginal tissue and 50 cases of different pathological types of human breast cancer and50 cases of lymph node metastases showed that there is a significant positive correlation between Pokemon expression and ER and PR in different pathological types of breast cancer and lymph node metastasis cancer tissues.(P < 0.05)Conclusion 1)Silencing Pokemon can inhibit the expression of ER and PR in breast cancer cells(MCF7 and T47D),which may be related to downregulation of ER expression by inhibiting ER promoter activity.2)There is a positive correlation between Pokemon expression and ER and PR in human breast cancer tissues and lymph node metastasis cancer tissues. |
