Objective : To explore the effect of Pokemon on TGF-β1 expression in human breast cancer cells and to analyze the clinical correlationship between Pokemon and TGF-β1.Methods: Experiment was divided into three parts. Part I: MCF-7 and MDA-MB-231 breast cancer cells were selected as cell models. The pokemon high expression cell model was obtained by transient transfection of PCDNA3.1/Pokemon.The mRNA expression levels of Pokemon and TGF-β1 were detected by Q-PCR analysis,and the protein expression levels of Pokemon and TGF-β1 were detected by immunofluorescence analysis.Part II: The Pokemon promoter activity was determined by Luciferase reporter assay Part III: To unveil the differential expression of Pokemon and TGF-β1,the tissue chip with 185 cases of breast cancer tissues, and immunohistochemistry staining method was utilized.Results: 1) Pokemon effect the expression of TGF-β1 in breast cancer cells.(1)Overexpression of Pokemon can inhibit the mRNA and protein level of TGF-β1 in MCF-7 cell.(2)Overexpression of Pokemon can inhibit the protein level of TGF-β1 in MDA-MB-231 cell. 2) The mechanism of Pokemon on TGF-β1 in breast cancer.(1)Overexpression of Pokemon can inhibit the activity of TGF-β1 promoter in MCF-7 cells.(2)Overexpression of Pokemon can inhibit the activity of TGF-β1 promoter in MDA-MB-231 cells. 3)The correlation between Pokemon and TGF-β1 expression in breast cancer tissues.Taken the 185 cases of human breast cancer tissues(including all kinds of pathological types) to immunohistochemical analysis, the expression of Pokemon and TGF-β1 was not significantly correlated, the difference between them was not statistically significant(p> 0.05).Conclusion:1)Pokemon inhibit the expression of TGF-β1 in breast cancer cells(MCF-7 and MDA-MB-231 cells), its mechanism might be suppress promoter of TGF-β1.2)The expression of Pokemon and TGF-β1 exhibit no sinificant correlation(r=-0.077, P=0.296) in human breast cancer tissues, when analyzed regardless of breast cancer types.
