ObjectiveAlzheimer’s disease(AD),is the most common cause of dementia,also known as dementia disease,is a progressive development of the nervous system degenerative diseases.Clinically,characterized by memory impairment,aphasia,loss,loss,visual impairment,executive dysfunction,personality and behavioral changes and other comprehensive dementia is characterized by the cause so far unknown.According to the International Alzheimer’s Disease Association’s "Global Alzheimer’s Disease Report 2015,,only about one year in 2015,about 10 million cases of Alzheimer’s disease,equivalent to about 19 people per minute Sick.The report predicts that by 2050,the number of patients with global Alzheimer’s disease will increase from the current 47 million to 132 million.At present,based on the huge population base,the number of AD patients in China ranks first in the world.This disease is not only tortured patients,returned to the family and society caused a heavy burden.Unfortunately,in the face of the ever-increasing incidence of the number of scientists on the etiology of this complex disease is still not very understanding,for the prevention and treatment of the disease has little effect.Professor Lai Shi long,founder of the Department of experimental data show that the party can not only alleviate the clinical symptoms of dementia patients,and can protect a variety of dementia animal model of learning and memory function.The mechanism of protective effect is mainly anti-A βtoxicity,inhibition of neuronal cell Ca2+ influx,anti-oxidative stress and anti-apoptotic,etc.,has great research value.However,the composition of traditional Chinese medicine compound is more complex,and its main effective drug composition is difficult to explain,so this study carried out an in-depth study of the party,the whole water decoction extraction,which acetyl ester extraction site has significant promotion of cell proliferation and Anti-A&toxicity.On this basis,we carried out the experimental study of the effects of Bushen Yizhiqifang extract on IBO induced dementia rat model.Purpose:BSYZ-E on the learning and memory ability of IBO-induced dementia rat model and its protective effect,hoping to help find the ideal method for the treatment of AD,for the further development and clinical use of this method Provide more effective experimental evidence.The focus of this experiment is to study whether the effective extraction of Bushen Yizhi No.3 can improve the symptoms and pathological changes of senile dementia by attenuating the excitotoxicity of IBO-induced rat hippocampus.Method:3-month-old SPF grade SD rats were randomly divided into 6 groups according to sex stratification,namely,blank control group,model group,positive drug group(donepezil),BSYZ-E low dose group,BSYZ-E middle dose group,BSYZ-E high dose group.Male and female half,given the corresponding liquid for 4 weeks.Morris water maze was used to detect the spatial learning and memory ability of rats in each group.(AchE)and acetylcholinesterase(ChAT)in hippocampus of rats were detected by HE staining.The expressions of BDNF,GDNF,NGF,SYT,SYP in hippocampus were detected by western blot and qPCR.,PSD95,TrkA,TrkB,TrkC and ERK1/2 mRNA and protein expression levels.The level of BDNF in hippocampus was detected by enzyme-linked immunosorbent assay(ELISA).Data were analyzed by SPSS17.0 statistical analysis software.Morris water maze positioning flight test escape latency was measured by repeated variance analysis.The other experimental data were analyzed by one-way ANOVA.LSD test was used to compare the two groups.P<0.05 was statistically significant.Result:1.Behavioral tests include Morris water maze and conditional fear experiments(1)Morris water maze behavioral resultsIn the BSYZ-E treatment of IBO induced dementia rats,the model group compared with the control group,the localization of navigation test escape latency extended(P<0.01),space exploration test in the target quadrant to shorten the time span across the platform But also decreased,suggesting that IBO-induced dementia model group of rats with impaired learning and memory.(2)The results of fear of experimental resultsIn the experiment of BSYZ-E treatment of IBO-induced dementia rats,the stagnation time of the model group was significantly shorter than that of the control group(P<0.05).Compared with the model group,the stagnation time of the positive drug Donepezil group and BSYZ group was significantly longer than that of the control group(P<0.05).The stagnation time of BSYZ medium dose group was prolonged,but the difference was not statistically significant(P>0.05)2.HE staining test results show that BSYZ-E and positive drugs(donepezil)on IBO dementia rat brain neurons have a certain degree of protection.3.The results of colorimetry showed that BSYZ-E could decrease AChE activity in hippocampus of IBO rats and improve ChAT activity in hippocampus of IBO rats.4.The results of Western-blot showed that BSYZ-E could increase the expression of BDNF,NGF,pCREB,SYT,SYP,PS095,TrkA,TrkB and ERK1/2 in the hippocampus of IBO rats.5.The expression of BDNF,GDNF,NGF,SYT,SYP,PS095,TrkA,TrkB,TrkC and ERK1/2 in the hippocampus of IBO rats were increased by the method of qPCR.6.ELISA kit showed that BSYZ-E could significantly increase the content of BDNF in hippocampus of IBO rats.Conclusion:Water maze test and conditional fear test results suggest that BSYZ-E can improve the learning and memory ability of IBO rats to a certain extent.BSYZ-E can improve the learning and memory ability of IBO rats by increasing the expression of BDNF,NGF,pCREB,SYT,SYP,PS095,TrkA,TrkB and ERK1/2 in hippocampus of IBO rats.The mechanism of BSYZ-E improving the learning and memory impairment of rats may be related to activation of BDNF/Erk/CREB signal.From the experimental results can be found BSYZ-E on Alzheimer’s have some therapeutic effect,it is worth further study.
