BackgroundAlzheimer ’ s Disease,also known as a common senile dementia,is for the memory deficit and related cognitive dysfunction as the main feature of neurodegenerative disease in the central nervous system.Along with the global aging process intensifing,it is expected that the whole world will approaching 115.4 millions of AD patients in 2050.Currently,drugs approved by FDA for the treatment of AD includes acetylcholine esterase inhibitors such as donepezil,galantamine and memantine as NMDA receptor antagonists.But these single target drugs can only improve some partial clinical symptoms,and can not slow down the progress of AD.To prevent and control AD,many scholars advocate that should be adopted comprehensive intervention of multiple targets.The prevention and treatment for disease,traditional Chinese medicine is a holistic concept and treatment based on syndrome differentiation as a guide,and the traditional Chinese medicine compound,is more compatible for use under the guidance of Chinese medicine theory,characteristics of action so it has multi targets and multi ways and multi links.Bushen yizhi formula is created by Professor Lai Shilong,earlier research showed that,the recipe not only can improve the clinical symptoms of dementia,the ability of learning and memory and can improve a variety of dementia animal models.The mechanism mainly for anti A beta toxicity,inhibiting cell influx of Ca2+,anti apoptosis.But the complexity of Chinese herbal compound composition,its main effective component is difficult to interpret,to conduct in-depth research on the formula decoction extraction,including acetyl ether extract can promote cell proliferation and anti Aβ toxicity.ObjectiveTo study the bushen yizhi decoction extracts three drug anti AD effect,we using sodium pentobarbital induced learning and memory impairment in mice model,its effect on the mice model of learning and memory behavior influence and its antioxidant effect.To provide more reliable experimental data for the further development of bushen yizhi recipe.MethodsMale Kunming mice were randomly divided into 5 groups,divided into blank control group,model group,BSYZ low dose group,BSYZ medium dose group,high dose BSYZ group,to give the corresponding by gavage for 4 weeks.Behavior using the Morris water maze,avoiding experiment to detect spatial learning and memory ability of mice in each group and passive escaping ability.To investigate the antioxidant effect,the SOD activity and the MDA content of cortex be detected.Datas are using SPSS17.0 for statistical analysis,the escape latency of Morris water maze navigation trials is using repeated measures analysis of variance.Other datas are using single factor analysis of variance,between the two two groups were compared with LSD test,there were statistically significant differences in P<0.05.Results1.Morris water maze testCompared with the normal group,model group,positioning navigation experiments in escape latency was significantly prolonged P<0.05,space exploration times to cross the target P<0.05 were significantly reduced;compared with the model group,low dose group and high dose group of navigation positioning the escape latency was significantly shortened P<0.05.2.Step-through testCompared with the normal group,model group,error frequency significantly increased P<0.05;compared with the model group,low dose group,middle dose group and high dose group the number of errors and model groups were statistically significant differences in P<0.05.3.The oxidative stress related indexOxidative stress related index results show that,compared with the normal group,model group,MDA significantly increased P<0.05 content,SOD activity significantly decreased P<0.01;compared with the model group,low dose group,SOD activity was significantly increased in the P<0.01 dose group,SOD activity significantly increased P<0.01,.SOD activity was significantly increased in the high dosage group P<0.05,middle dose group have significant differences in P<0.01.ConclusionBSYZ extracts can improve the learning and memory impairment caused by pentobarbital sodium in mice,and improve the behavior performance in Morris water maze and Step-through test.BSYZ extracts can improve the SOD activity in the mouse cerebral cortex with the anti-oxidation effect,it is value to further research for its treatment effect for AD. |
